| Objective:To investigate the expression of special AT rich sequence binding protein 1 and breast cancer metastasis suppressor 1 protein in salivary adenoid cystic carcinoma tissues,pleomorphic adenoma tissues and normal salivary tissues,and to find out their role and clinical significance in the occurrence and development process of salivary adenoid cystic carcinoma.Methods:50 cases of adenoid cystic carcinoma,50 cases of pleomorphic adenoma and 50 cases of normal salivary tissues(>1.5 cm away from the cancer tissues)were collected from Yantai Yuhuangding Hospital,Q ingdao University.The expression of SATB1 and BRMS1 protein were detected by the immunohistochemical method.To compare the differences of the expression of the two proteins in the three tissues.To analyze the relationship between SATB1 and BRMS1 protein and the clinical and pathological features,such as the patients’ age,gender and degree of differentiation,T stage,pathological type,distant metastasis of adenoid cystic carcinoma,and to analyze the correlation of SATB1 and BRMS1 protein expression in adenoid cystic carcinoma.Results:(1)The expression of SATB1 protein is the highest in adenoid cystic carcinoma tissues(44%),while the lowest in normal salivary tissues(12%),and middle in pleomorphic adenoma tissues(18%).The difference of the groups has statistical significance(χ2=15.570 P<0.01).But,the expression of BRMS1 protein is the lowest in adenoid cystic carcinoma tissues(36%),while the highest in normal salivary tissues(72%),and middle in pleomorphic adenoma tissues(62%).The difference of the groups has statistical significance(χ2=14.063 P<0.01).(2)The positive expression rate of SATB1 protein in poorly differentiated adenoid cystic carcinoma tissues was 66.67%,which was significantly higher than the well-differentiated and moderately differentiated tissues(27.59%,P<0.05);In the pathological type,the positive expression rate of solid type tumor tissues was about 65.00%,which was significantly higher than the sieve/ tube type(30.00%,P<0.05).The positive rate of distant metastasis group was about 78.57%,which was significantly higher than the non-metastasis group(30.56%,P<0.05).The expression of SATB1 protein was closely related to the degree of differentiation,pathological type,and distant metastasis of adenoid cystic carcinoma,but not have statistical significance with patients’ age,gender and T stage of adenoid cystic carcinoma.(3)The positive expression rate of BRMS1 protein in poorly differentiated adenoid cystic carcinoma tissue was 19.05%,which was significantly lower than the well-differentiated and moderately differentiated tissue(48.28%,P<0.05);The positive rate of distant metastasis group was about 14.29%,which was significantly lower than the non-metastasis group(44.44%,P<0.05).The expression of BRMS1 protein was closely related to degree of diffetentiation and distant metastasis of adenoid cystic carcinoma,but have no statistical significance with patients’ age,gender,and T stage,pathological type of adenoid cystic carcinoma.(4)SATB1 and BRMS1 protein expressed in 7 cases of adenoid cystic carcinoma tissues.But,neither SATB1 nor BRMS1 protein expressed in 17 cases of adenoid cystic carcinoma tissues.The expression of SATB1 protein had no correlation with BRMS1 protein in adenoid cystic carcinoma tissues(γ=-0.077,P=0.594).Conclusion:(1)The expression of SATB1 protein in adenoid cystic carcinoma tissues was higher than that in pleomorphic adenoma and normal salivary tissues.The expression level of SATB1 protein is related to the differe ntiation,pathological type and distant metastasis of the tumor.(2)The expression of BRMS1 protein in adenoid cystic carcinoma tissues was lower than that in pleomorphic adenoma and normal salivary tissues.The expression level of SATB1 protein is related to the differentiation and distant metastasis of the tumor.(3)There was no correlation between SATB1 and BRMS1 protein expression in adenoid cystic carcinoma.(4)SATB1 and BRMS1 may be closely related to the occurrence and development of SACC.SATB1 and BRMS1 have no direct regulation relationship in the occurrence and development of adenoid cystic carcinoma.Combined detection of the two protein can provide a theoretical basis for the clinical diagnosis and the prognosis evaluation of adenoid cystic carcinoma.SATB1 and BRMS1 are expected to become new targets for gene therapy of adenoid cystic carcinoma. |
