Objectives :To investigate whether dual specificity phosphatase 3(DUSP3),Arf GAP with SH3 domain,ankyrin repeat and PH domain 1(ASAP1),guanosine binding protein5(GBP5)gene in tuberculosis patients and tuberculosis latent population.Through the expression of DUSP3,ASAP1,and GBP5 genes in patients with tuberculosis and latent tuberculosis infection,the differences in gene expression between patients with confirmed tuberculosis,latent tuberculosis infection,and non-tuberculosis population were explored.It has targeted different groups to develop different prevention and treatment programs.Methods:The patients admitted to The Affiliated Hospital of Qingdao University and Qingdao Huangdao District Tuberculosis Hospital from August 2016 to May 2017 were selected as the study subjects and the health examination personnel of the Health Examination Center of the Affiliated Hospital of Qingdao University were the subjects of the study.Whether sputum smears were found to be Mycobacterium tuberculosis(MTB)and Interferon-? release tests(IGRAs)as the criteria,the eligible subjects were divided into 3 groups,of which 40 cases were diagnosed with pulmonary tuberculosis,tuberculosis latent 40 cases in the infection group and 40 cases in the healthy control group.3 ml of peripheral venous blood in the three groups of subjects were extracted with lithium heparin anticoagulation test tubes to extract human peripheral blood mononuclear cells in the blood,and then to extract RNA from them and reverse transcriptase synthesis.c DNA,using real-time PCR to detect the gene expression of these three groups of people,compared the relative expression of these three genes 2-??Ct.SPSS software was used to test the homogeneity of variance before data analysis,P> 0.05,which means that the variance is equal,the ages of the three groups are denoted by x ±s.Graph Pad Prim 5.0software is used for statistical analysis of the data,T test is used to compare the mean of the two groups,the mean difference between the three groups is compared,P<0.05 The difference was statistically significant.Results : GBP5 gene expression in the three groups of subjects was statistically significant(F = 7.23 P = 0.001).The relative expression of gene in tuberculosis group was significantly higher than that in latent infection group(1.58±0.80 vs 1.09±0.68),the difference was statistically significant(t =2.93,P=0.004),the relative expression of the confirmed tuberculosis group was significantly higher than that in the healthy control group((1.58±0.80 vs 1.04±0.61),the difference was statistically significant(t = 3.40,P<0.01).There was no significant difference between the tuberculosis latent infection group and the healthy control group(1.09±0.68 vs 1.04±0.61;t = 0.39,P = 0.70).There was no significant difference in the gene expression of ASAP1 gene among the three groups(F =0.26 P=0.77).There was no significant difference in the gene expression of DUSP3 gene between the three groups(F = 2.21 P = 0.11).Conclusion:1.The expression of GBP5 gene in tuberculosis confirmed tuberculosis latent infection group,healthy control group there are differences.2.There was no significant difference in the expression of DUSP3 in the three groups of people,but the small sample size and the possibility of error were not excluded.3.There was no significant difference in the expression of ASAP1 in the three groups of people,but the small sample size and the possibility of error were not excluded.4.There are differences in gene expression of GBP5 gene in tuberculosis patients,tuberculosis latent infection populations and healthy people.The GBP5 gene can screen for tuberculosis patients,which has important guiding significance for the diagnosis,treatment and prevention of tuberculosis.For latent TB-infected people,the difference in expression between the occult group and the healthy control group was not confirmed.
