Objective: To explore Caprin-1 expression in clinical glioma specimens and its influence on biological characteristics of glioma cell line,providing the theoretical basis for gene targeting therapy and evaluating the prognosis of glioma.Methods: Thirty-one clinical specimens were collected and the relationship between expression of Caprin-1 and tumor grading was analyzed.With U251 cells as the experimental object,Caprin-1-overexpressed U251 cell line was established.The experiment was composed of three groups: the blank control group(wild-type U251 cells),the negative control group(U251 cells with pEGFP-C1 plasmid),and the experimental group(U251 cells with pEGFP-C1-caprin-1 plasmid).U251 cells were transfected with Caprin-1-containing expression vector using lipofectamine3000,and the expression level of Caprin-1 mRNA and protein was detected by RT-PCR and Western blot in three groups of cells.Cell proliferation and migration ability were determined with Real-time cell analyzer,enhanced CCK-8 kit,scratch assay and Transwell test respectively.Results: The results of immunohistochemistry showed that the expression of Caprin-1 in glioma was located in cytomembrane and cytoplasm,and its expression increased with the malignant level of tumor tissue(P?0.05).In cell experiment,the results of RT-PCR and Western blot showed that the expression protein levels of Caprin-1,p-Akt,CyclinD1 and the expression mRNA level of Caprin-1 in the experimental group were higher than those in the control groups(P?0.05),and the experimental group displayed faster proliferation rate determined by real-time cell analyzer and CCK-8 than the controls(P<0.05)while there was no significant difference between the two control groups(P>0.05).The results of the scratch assay and Transwell experiments showed higher migration ability compared with the two control groups(P?0.001).Conclusion: Our experiments demonstrated that the expression of Caprin-1in glioma was increased with the malignant level of tumor tissue.The proliferation rate and migration ability of U251 cells were elevated by the overexpression of Caprin-1.
