| Objective:To observe the optoacoustic/ultrasonic imaging effect and safety of indocyanine green and liquid perfluorohexanes loaded dual-modal photoacoustic/ultrasound nanoparticles contrast agents decorated with SDF-1 in the lymph node metastasis of rabbit with tongue cancer.Methods: Preparation of the nanoparticles agent,PLGA shell,ICG and PFH kernel,decorated with SDF-1 by means of emulsification technique.To observe the dual-modal photoacoustic/ultrasound optoacoustic imaging effect in the lymph node metastasis of rabbit with tongue cancer,the photoacoustic intensity of different contrast agents were compared at the same time.After 1 weeks of injection of contrast agent and,all the healthy rabbits underwent the physical examination,complete blood count,liver and renal function tests.During this period,the emotion state and vital signs of rabbits should be evaluated.The rabbits were killed after one month,and the pathological examination of the irradiated and non-irradiated skin,lung,liver and kidney were carried out.Results:After the injection of indocyanine green and liquid perfluorohexanes loaded nanoparticles contrast agents decorated with SDF-1,photoacoustic imaging began to occur in 20 mins and disappeared in 24 h.The effect of photoacoustic imaging of metastatic lymph nodes was significantly enhanced by indocyanine green and liquid perfluorohexanes loaded nanoparticles contrast agents decorated with SDF-1,Little photoacoustic signal was found in the blank group.After the contrast laser irradiation was performed in the ultrasound experimental group,the ultrasonic imaging of lymph nodes in the neck was gradually increased,and the peak was reached in about 30 minutes.Compared with the ultrasonic control group,the imaging enhancement was more obvious.There was no significant difference in physical sign between the experimental group and the control groups during the first month after intravenous injection of contrast agent.The histological examination of liver、lung 、kidney and the skin of irradiated area was not significantly different from that of the normal rabbits.Conclusion: Indocyanine green and liquid perfluorohexanes loaded nanoparticles contrast agents decorated with SDF-1 can significantly enhance the photoacoustic/ultrasound imaging of metastatic lymph nodes.The results of this study indicate that the agents is a safe,effective and inexpensive synergist. |
