【Object】 Embryo implantation is a complex process involving synchronized crosstalk between a receptive endometrium and a functional blastocyst.Apoptosis,is a death process that can maintained the homeostasis of the internal environment and can be better adapt to the living environment.Apoptosis plays an important role in embryo implantation and maintenance of pregnancy.On the time of the implantation of the embryo,endometrial epithelium is rapidly apoptotic,so that the embryo can be implanted smoothly.After the implantation,the apoptosis of the decidua have a vital role in the process of implantation and the successfully pregnancy.Progressed cell death 4(Pdcd4)is a highly conversed protein that can inhibit the occurrence,development,and invasion of tumor through suppress the transcription and translation of genes.The previous results conducted by our group demonstrated that the expression of PDCD4 increased from DI-D4 and decreased from D5-D7.Besides,the expression level in implantation sites(IS)is lower than that at interimplantation sites(IIS).It also demonstrated that the level of PDCD4 in clinical endometrium samples of threatened abortion and missed abortion is significantly higher than normal group.So,in this search,we analyzed the effect of over expressed PDCD4 in mouse ESCs on the apoptosis and decidualization.【Methods】(1)Constructed the overexpressed plasmid vector(p CMV6-Pdcd4/p CMV6-vector)and amplification and extraction in Escherichia coli.The PDCD4 over expressed in ESCs via the cell nuclear membrane transfection technique.The expression level of PDCD4 is higher expressed in treatment group.(2)Analyzed the effect of the over expressed PDCD4 on ESCs’ apoptosis and decidualization by using real-time quantitative polymerase chain reaction(q PCR),Western blotting(WB)and flow cytometry.【Results】(1)The expression level of PDCD4 in treatment group is significantly higher than that in control group,the endometrial primary stromal cell overexpression model was successfully established.(2)After the overexpression of PDCD4,the apoptosis rate of ESCs is increased and the expression levels apoptosis related factors and proapoptotic factors in intrinsic or mitochondria-associated pathway were increased.(3)Overexpression of the Pdcd4 gene could inhibit the establishment of decidualisation.For the maintenance of the decidua,a decrease in the expression levels of Dtprp was observed,but this trend was not statistically significant.【Conclusion】The overexpression of PDCD4 is detrimental to the establishment and development of decidua via increasing apoptosis of mouse endometrium stromal cells,which indicated that the abnormal expression of PDCD4 have the vital role in embryo implantation.However,its specific mechanism still needs to be further studied.
