| Objective:To explore the clinical features,efficacy,and determinants on transformation in patients with Idiopathic cytopenia of undetermined significance(ICUS).Methods:The clinical and pathological features were analyzed retrospectively in 54 patients with ICUS,and 25 common genes in patients with MDS/MPN were detected with Next-generation DNA sequencing.Results:The efficacy rate of treatment was 35.2% in all 54 patients.Gene mutations significantly affect the treatment effect and clinical transformation.A total of 31.5% of patients had at least one mutation.The most common mutations were TET2,followed by DNMT3 A,ASXL1,and SRSF2.Conclusion:The clonal hematopoiesis of somatic mutation may be an important factor affecting the therapeutic effect and clinical transformation in ICUS patients,and DNA sequencing should combine with morphology and cytogenetics in order to achieve the final diagnosis and predict disease progression. |
