Protective Effects Of Huayu Tongluo Traditional Chinese Medicine On Diabetic Nephropathy Rats And On The Expression Ofsignal Transduction Pathway Of PI3K/Akt

Objective: This experiment study protective effects of Huayu Tongluo medicine role,and explored its possible mechanism.Methods: 85 healthy male Sprague Dawleg rats were randomly divided into control group(10 rats),model group.The normal group was fed with ordinary feed and the moulding group was fed with high sugar and fat feed.And the end of six weeks,the model group were established with high sugar highfat diet combined with stretozotocin(35mg/kg)by once intraperitoneal injection.After successful modeling,rats were randomly divided into model group,Huayu Tongluo group,the Irbesartan group,Huayu group and Tongluo group,were 15 and Huayu Tongluo group were 13.Different groups were fed corresponding drugs once a day for 16 weeks.According to the dosage of Chuanxiong,1.13 g/kg,Danshen1.41g/kg,Dilong0.94 g/kg,Shuizhi and Quanxie 0.56 g/kg,and the erbesartan 14.12mg/kg filled the stomach of rats.At the end of the 16 th week,hour urine,body weight,water quantity,food intake,24 h urinary total protein and renal function,blood glucose,blood lipids were tested.Pathomorphology changes of kidney were observed with light microscope and electron microscope.The experssion of PI3 K was detected by Immunohistochemical staining respectively staining and Real-time PCR.Results:1.The general status of ratsThe rats in normal control group were in good condition and flexible activity.Food intake,water intake and urine output were abnormal;each group modeling of food intake,water intake and urine output wereabnormal,and weight increased slowly.The rats has slow reaction and movement,and listless spirit.The model group were the most obvious.2.The biochemical indicators of ratsThe results show that compared with the normal control group,each group modeling water quantity,food intake,24 h urinary total protein were significantly increased(P<0.05);fasting blood glucose were significantly increased(P<0.05);model group was significantly higher in Scr level(P<0.05)and in BUN level(P<0.05)and in TC,TG,LDL levels(P<0.05).With the model group,each treatment group water quantity,food intake,24 h urinary total protein were significantly decreased(P<0.05);fasting blood glucose were significantly decreased(P<0.05);Each treatment group was significantly lower in Scr level(P<0.05)and in BUN level(P<0.05)and in TC,TG,LDL levels(P<0.05).Each treatment group were obviously increased LDL level(P<0.05).Other groups were significantly reduce-d,and the Hauyu Tongluo group is most obvious.3.Morphological observation of kidneyLight microscope observation :normal control group rats had clear and normal kidney tissue structure;model group rats glomerular hypertrophy,renal capsule was slit-like even balloon adhesions,thickening of the basement membrane,increasing mesangial matrix;compared with the model group,other groups were significantly reduced pathological changes.Electron microscope observation: model group rats heterogeneity of glomerular basement membrane thickening,increased mesangial matrix,podocyte foot processes were Irregularly fusion;compared with the model group,the pathological changes in other groups significantly reduced.4.Immunohistochemical and Real-time PCR detection results of the expression of PI3K、Akt in kidneyPI3K was mainly expressed in nucleus,PTEN 、 Akt1 was mainly expressed in Cytoplasm.Compared with the normal group,the expression was significantly increased in model group(P<0.05);compared with the model group,The expressions of PI3 K and Akt1 in each treatment group weresignificantly decreased(P<0.05),and the expressions of PTEN were significantly increased(P < 0.05).Conclusions:1.Huayu-Tongluo TCM can improve the general status of DKD rats,lower total 24-hour urine protein,improve lipid metabolism disorder and lower Scr and blood urea nitrogen levels,has a protective effect in the DN rats kidney.2.Huayu-Tongluo TCM can improve the symptoms of multi-drink,multi-food and urine,improve lipid metabolism disorder,and reduce24 UTP.The combination of Huayu-Tongluo TCM is better than Huayu TCM and Tongluo TCM.3.Huayu-Tongluo TCM can reduced PI3 K 、 Akt 1expression levels,increased expression levels of PTEN.Huayut-ongluo TCM may be another way to regulate this pathway to prevent foot cell injury and reduce proteinuria.
