Objective:Based on the theories that the pyramidal tracts are aggregations of efferent nerve fibers from the upper motor neurons and are involved in the control of motor functions of the body,the possible cause of the development of SCP(spastic cerebral palsy),neurotoxicity of alcohol,the pyramidal tracts of adult rats have been located and injected with alcohol,followed by examinations of animal ethology and pathology,in order to make a stable and reliable animal model of SCP.Methods:Thirty-six adult SPF-grade SD rats,half male half female,weighing 220g-250 g,provided by the Animal Experimental Center of Hebei Medical University,raised under the barrier environment of Experimental Animal Center of the Third Hospital of Hebei Medical University were involved and divided into control group A,experimental groups B and C according to the random number table method.After the proceed of craniotomy and localiz-ation of pyramidal tracts,C were injected with absolute alcohol,B physiological saline and A nothing.Animal motor tests including involuntary movement,muscular tension test,ambulation,hindlimb foot angle,surface righting test,ramp test,hindlimb suspension test,inclined plate test,open field test,resist arrest test were observed on the 28 th day after surgery in each group of rats.After all,pathological examination of brain tissue sections was performed to observe whether pathological changes occurred,such as cell edema,irregular arrangement of cells and organelles,necrosis and so on.Results:1.The survival rate of A was 100.0%(12/12),B 91.7%(11/12)and C 75.0%(8/12).2.There was no statistically significant difference in body weight between the three groups until 3-day post-operation.Since then,the weight gain of C had increased slowly compared with others.3.In all motor tests above,statistically significant difference existed in C vs A and C vs B.Obviously rats in C encountered with problems like continuous spasticity of right limb,increasing muscular tension,involuntary movements,larger hindlimb angles,poor flexibility,impaired body balance,partial anticlockwise circling movements,low excitability,poor learning ability and memory.4.Pathological changes like cell edema,irregular arrangement of cells and organelles,necrosis were found in examination of brain tissue sections of C,not A or B.Conclusions: Injection with absolute alcohol into the pyramidal tracts of adult rats can make scientific,stable and repeatable animal model of SCP.
