Objective:To establish the synthesis of fully automated one-step 11C labeled paraquat,and to obtain the pharmacokinetic information of[11C]paraquat in the rabbit.Methods:Using the TRACERlab FXC synthesis platform,under 80?reaction conditions in alkaline conditions[11C]CH3I reacted with acetaz-olamide.Then using alcohol as eluent,through high performance liquid chromatography?HPLC?,[11C]paraquat was isolated from reaction solution.After intraperitoneal anesthesia of the rabbit,[11C]paraquat was injected into rabbit via auricular vein of the rabbit.Then the VIP scanning model of GE discovery elite PET/CT was used for continuous dynamic scanning at0min,5min,10min,20min,30min,40min.Results:The synthesis of the agent was about 70%,the specific activity was 210-1500Ci/mmol,the time for the whole producing process was only about 30 min.PET and PET/CT was used to scan the rabbit with[11C]paraquat intravenously.Then the scanning images of PET and PET/CT were fused and found that[11C]paraquat was rapidly distributed in the rabbit,and the brain?basal ganglia?,lung,liver?gallbladder?and kidney showed continuous uptake and the kidney was barely excreted in the scanning time range.Conclusion:[11C]paraquat can be used as an imaging agent to study the biodistribution of paraquat in living tissues and cells after paraquat poisoning,and to study the mechanism of paraquat poisoning.
