Objectives: Study the changes of serum IL-2,IL-10 and TNF-ain mice with ulcerative colitis(UC)under high-fat diet,to explore the mechanism of high-fat diet on UC.Methods: 80 healthy male Balbc mice were randomly divided into control group,high-fat diet group,DSS group,DSS+high-fat diet group, with 20 mice in each group.Acute and chronic UC model were induced by freely drinking of 3% DSS solution. The inflammatory degree of mice was judged by disease activity index score through the changes of diarrhea, hematochezia and body weight.Colon tissue was observed under light microscope, colon injury was judged by histopathological score.Levels of serum il-2,il-10 and TNF-a were detected by ELISA.Results: the scores of histopathological and disease activity index in DSS + high fat group were significantly higher than DSS group(P < 0.05).TNF-a level in DSS + high fat group was significantly higher than DSS group, while Il-2 and Il-10 levels in DSS +high fat group were significantly lower than DSS group(P< 0.05)Conclusions: high-fat diet could obviously aggravate colitis, the possible mechanism was that the immune disturbance was aggravated by increasing TNF-a level and decreasing Il-2 and Il-10 levels.
