Hypopharyngeal carcinoma is one of the most malignant tumors of head and neck squamous cell carcinoma.It is prone to local metastasis and distant distant spread,and the prognosis is poor.Due to the hidden location of the disease,most patients are diagnosed in the late stage.In clinical surgery is the first choice,sometimes combined with adjuvant chemotherapy and radiotherapy,however,the five-year survival rate is still low and with a downward trend.With the increasing emphasis placed on the research of cancer treatment by Chinese medicine in recent years,we hope to find a new approach to treating head and neck cancer by traditional Chinese medicine so as to improve the patient’s survival rate and quality of life.To this end,we chose the common Chinese medicine Actinidiachinensis planch as our research drμg.Objective:To investigate the inhibition efficacy of Actinidiachinensis planch extracted by ethyl acetate(EE-ACP)on human cancer cell line Fadu cells’ proliferation and induction on apoptosis,and to explore its potential mechanism.Methods:Using the CCK8 assay,Hochest33258 stain,flow cytometric(FCM)analysis and Western Blot methods to investigate the EE-ACP’s affects on Fadu cells and its potential mechanism.Results:1.CCK8: The result of CCK8 showed that,after 100μg/ml、200 μg/ml、300 μg/ml、400 μg/ml、600 μg/ml、800 μg/ml、1000 μg/ml、1250 μg/ml EE-ACP treatment for 24、48、72 h,the proliferation of Fadu cells were obviously inhibited in a dose-and time-dependent manner.The IC50 was300,160 and 100 μg/ ml.2.Hoechst33258: The result of hoechst33258 showed that,after treated by 150 μg/ml、250 μg/ ml、350 μg/ ml、450 μg/ ml、550 μg/ ml EE-ACP for 48 h,the apoptosis morphology of Fadu cells was observed gradually increased with the increase of drμg concentration.3.Flow cytometric(FCM)analysis : The apoptotic Fadu cells were observed after treated with 50 μg/ ml、100 μg/ ml、200 μg/ ml 、300 μg/ ml、400 μg/ ml EE-ACP for 48 h and the effect was enhanced as the dosage increased.4.Western blot:The results showed that with the increase of drμg concentration,the expression of P53 protein gradually decreased.ConclusionEE-ACP inhibits the proliferation of Fadu cells,probably throμgh the down-regulation of P53.
