Objective: With the acceleration of the aging trend of the international population,it is one of the hot topics in the medical field to explore the mechanism of aging and anti-aging.Although aging is not explicitly defined as a disease,there is no clear boundary between aging and the pathophysiological changes of many elderly diseases.Without appropriate intervention,it leads to the occurrence of geriatric diseases.In this experiment,Coccomyxa gloeobotrydiformis(CGD)is water extract of gibadeceae,a kind of micro-algae-green algae.In our previous study,we found that CGD not only can reduce the focal cerebral ischemia-reperfusion injury and improve the learning and memory ability of aging rats,but also significantly improve the metabolic syndrome rat model of sugar,fat metabolism.In this study,we will explore the anti-aging effects of CGD under different dietary conditions in combination with the influence of metabolic factors,and further clarify its intrinsic mechanism.Methods:(1)48 male 10-month-old Balb / c male mice were randomly divided into six groups with 8 rats in each group: high calorie diet + NS control group,high calorie diet + CGD treatment group,diet restriction + NS control group,Diet restriction + CGD treatment group,normal diet + NS control group,normal diet + CGD treatment group.After grouping,each group was given the corresponding dietary conditions and given the same dose of Normal Saline(NS)or CGD orally for 15 weeks.(2)The general condition of mice in each group were observed dynamically.The body weight,abdominal circumference and metabolism related indexes such as blood glucose and blood lipid(TC,TG)were measured every 5 weeks.Fasting serum insulin,HDL-C and MDA were measured at the end of treatment.After the above experiment ended,the survival of each group of mice were recorded,sacrificed,drawn for subsequent experiments.(3)Coronal,liver,kidney,spleen,skeletal muscle and testis were fixed and embedded into paraffin sections.H & E staining was performed to observe the pathological changes of the above tissues.The heart,liver and kidney of important organs were aged Marker lipofuscin staining,observation of aging.(4)Western blot was used to detect the expression of p53,p16,p21,SOD1 and SOD2,the expression of autophagy-related protein LC3 B,the expression of TNF?in heart,liver and gastrocnemius tissues Express the situation.Results:1.CGD improved the general state of aging mice under different dietary conditions.2.CGD regulated the metabolism of old mice under different dietary conditions.(1)FBG,FINS and IRI decreased significantly after CGD treatment in high-calorie diet group and normal diet group,and there was no obvious change in FBG,FINS and IRI after CGD treatment in diet-restriction group;TG decreased significantly in CGD-treated group C significantly increased,high calorie diet group and dietary restriction group CGD treatment,TG,HDL-C changes were not obvious;under the same diet CGD treatment group was significantly less than the control group TC,body weight and abdominal circumference no significant difference.(2)Body weight and abdominal circumference in CGD treatment group were significantly higher than those in normal diet group and highcalorie diet group.The FBG in the diet-restriction group was significantly lower than that in the normal diet group and the high-calorie diet group,and there was no significant difference between the high-calorie diet group and the normal diet group.FINS and IRI in high-calorie diet group were significantly higher than those in normal diet group and dietary restriction group,and ISI was lower than normal diet group and dietary restriction group.There was no significant difference between TC and TG in the diet restriction group and the normal diet group,but TC in the high-calorie diet group was higher than that in the normal diet group,TG was significantly lower than the normal diet group and dietary restriction group.High-calorie diet group,diet restriction group,normal diet group HDLC no significant difference.P <0.05,with statistical significance.3.CGD reduced the histopathological changes in aged mice and reduced the formation of lipofuscin in heart,liver and kidney.4.The anti-oxidative effects of CGD: CGD decreased MDA in normal diet group,while CGD treatment in normal diet group,high calorie diet group and diet restriction group increased SOD1 significantly.P <0.05,with statistical significance.5.The proteins expression of CGD changed: CGD increased the protein expression of p53,p16 and p21 in the high-calorie diet group,the diet-restricted group and the normal dietary heart;the expression of LC3 B protein increased significantly after the CGD treatment in the high calorie diet group and the normal diet group.There was no significant change in LC3 B in diet restriction group.After CGD treatment in normal diet group,the expression of TNF?protein in heart and liver tissue was significantly decreased,while the change in liver tissue was not obvious.P <0.05,with statistical significance.Conclusion:1.CGD delays the aging process of aging mice under different diet conditions,of which the normal diet group and high-calorie diet group best.2.CGD significantly improves the normal diet group and high-calorie diet group of old mice glucose and lipid metabolism.3.CGD plays an anti-aging role through anti-inflammatory,anti-oxidative and autophagy levels,and its mechanism may be related to the regulation of p53 / p16 / p21 signaling pathway.
