Objective: This study observes the effects of icariin on repairing of alveolar bone defects in SD rats,and provide a more economical bone growth factor for the treatment of repairing of clinical alveolar bone defects.Methods: 1.The effect of Icariin on repairing of alveolar bone defects in SD rats.33 healthy 6 to 7-week-old male SD rats were selected and divided into 3 groups randomly(blank group,AGS group and ICA/AGS group).Establishing a 4mm×2mm×1.5mm box-type periodontal bone defect in the lower left anterior tooth of rats with hand-held craniocerebral drilling,then did treatment according to the groups:the blank group did suture directly,AGS group filled with AGS in the defect then suture,ICA/AGS group filled with ICA/AGS then suture.At 2W?3W?4W after operation,3 rats were sacrificed randomly in each group.Intercepted bone fragment and collected them,then performed by Micro-CT scanning,analyze the Bone volume/Tissue volume(BV/TV)?Trabecular number(Tb.N)?Trabecular Separation(Tb.Sp)?Trabecular Thickness(Tb.Th)of newborn periodontal bone in each group.Each bone fragment was sliced and stained with HE staining,to observe the periodontal tissue change condition.2.The best concentration of ICA to promote alveolar bone repair.66 healthy 6 to 7-week-old male SD rats were selected and divided into 6 groups randomly.Establishing a model of periodontal bone defect in rats with the same way as the first part,filled with different concentrations ICA/AGS according to the groups(1mg/m L?1.5mg/m L?2mg/m L?2.5mg/m L?3mg/m L in turn).At 2W?3W?4W after operation,3 rats were sacrificed randomly in each group.The specimens were collected and examined in the same way as the first part.Results: 1.The effect of Icariin on repairing of alveolar bone defects in SD rats.At 2W after operation,no significant new bone formation was observed in blank group and AGS group during the Micro-CT scan,however,a small amount of new bone was observed in ICA/AGS group.Three groups were observed formation of new bone after operation for 3W and 4W.Compared BV / TV,Tb.N,Tb.Sp,Tb.Th between the there groups at different time,experimental group was the best,which was significant difference from the other two groups(p <0.05).HE staining showed that bone repair was performed in all groups at 2W.At this time,hemorrhage and inflammatory granulation tissue were observed in the defect area.After operation for 3W,defect area was almost filled with fibrous tissue,the osteoblasts started to aggregate,and the osteogenesis is active in ICA / AGS group.The new trabecular bone was seen in all three groups at 4W,and the bone in ICA / AGS group was more mature.2.The best concentration of ICA to promote alveolar bone repair.After Micro-CT scanning and reconstruction,new bone formation was observed in all groups.Compared BV/TV,Tb.N,Tb.Sp,Tb.Th among each groups in different times,there was no significant difference at 2W.But the effect of the five groups were better compared with the blank group at 3W and 4W(p<0.05).Compared the indexes of groups A,B,D,E and C,only the bone mass of group A and C were statistically different at 2W(p<0.05),and the amount of bone mass and the quality of bone formation of group C showed better results at 3W.At 4W,there was a significant difference in the amount and quality of new bone between group A and group C,and the other three groups only had a significant difference in osteogenic quality compared with group C(p<0.05).Histological examination found that hemorrhages or new blood capillaries appeared in each group at 2W.At 3W,the vascular organized,the fibrous connective tissue appeared,and osteoblasts were aggregated with the appearance of new trabecular bone.After operation for 4W,the new newborn trabecular almost mature.Conclusion: ICA can promote the repair of alveolar bone defect in SD rats;2 mg /m L is the best when used alone in SD rats' intraoral environment.
