Lectin is a protein that promotes cell agglutination and is widely found in plants,animals and microorganisms.Leguminous plants have the most abundant lectin,consisting of two or four monomers,each with a molecular weight of about 25 ~ 30 KD,and a binding sugar site.The binding specificity of lectin in plants determines its many advantages in related functions and properties.In this study,Red kidney bean lectin(RKBL or phytohem-agglutinin,PHA)was extracted from yunnan real estate Red kidney bean.Existing studies have shown that Kidney bean lectin(KBL)can safely and effectively prevent fertilization,terminate pregnancy at different times and fight HIV.It is possible to develop a microbicide for the vagina or rectum that is both contraceptive and sexually transmitted,including HIV.As a long-term drug,its safety is particularly important.Therefore,we need to know about the carcinogenicity and anti-tumor properties of KBL,but there is little or no literature available.Therefore,this dissertation mainly discusses the red kidney bean plant lectin(RKBL)as the safety of long-term use of drugs,carcinogenicity,as targeted drug delivery carrier is feasible and inhibit embryo implanted around the time of implantation related pathways and mechanism of a preliminary study.The methods of this study were: 1.The female rabbit and the male rabbit and the male rabbit respectively carried on the vagina and the rectum to inject the red kidney bean agglutinin(RKBL)gel and the nonyl alcohol ether gel.The female rabbits were injected with 2ml /30 mg per vaginal injection,and the male rabbits were injected with4 ml /60 mg of red kidney bean agglutinin gel.2.RKBL on B16 melanoma cells and the influence of human liver cancer cells QGY-7701 morphology,determined by MTT method is used to detect RKBL on B16 melanoma cells and the proliferation of QGY-7701 cells,FITC-RKBL preparation and effect on tumor cells.3.The carcinogenicity of RKBL was studied by conducting vaginal or rectal continuous administration of female rats.4.By comparing the expression of STAT3 transcriptional factor STAT3 in pregnant mice treated with RKBL and control mice,a preliminary study was conducted to investigate the related pathways of RKBL in influencing the implantation of blastocyst and termination of pregnancy.Results: experiment 1: RKBL gel was used to maintain complete structure of female rabbit vagina and male rabbit rectal mucosal epithelial cells.However,the female rabbit vagina and the male rabbit rectal mucosal epithelial cells showed different degrees of breakage.Experiment 2: Red kidney bean plant lectins(RKBL)has inhibitory effect on tumor cells,and the tumor cell surface contains a large amount of KBL or PHA receptor,RKBL can induce tumor cell apoptosis and apoptosis occur.Experiment 3: The experimental results showed that the long-term vaginal supply of red kidney bean was safe for 18 months,which had no effect on the carcinogenicity of mice.Compared with the control group,the organ coefficient,blood blood,liver and kidney function of mice were compared with the control group,although the individual indexes showed significant differences,but they were fluctuated within the normal reference range.Experiment 4: The expression of STAT3 in the endometrium of pregnant mice injected with RKBL was inhibited,and the expression of endometrial STAT3 in high-dose group was decreased with the increase of time after injection.Conclusion: 1.RKBL gel can not cause damage to epithelial cells in female rabbit vagina and male rabbit rectum,and it has good prospect in developing new microbicide.2.RKBL has inhibitory proliferation and lethal effects on tumor cells,and the tumor cell surface contains a large number of RKBL receptors,which provides a strong basis for RKBL as a targeted drug.3.Long-term injection of RKBL was safe and did not cause cancer in the 18-month carcinogenic test.The coefficient of spleen of female rats in high-dose group was higher or lower than that in the control group.The adrenal coefficient and bladder coefficient were significantly higher than those in the control group.The adrenal coefficient of low dose group was significantly higher than that in the control group.In the high-dose group,the medium dose group and the low-dose group,the coefficient of the organ was different from that of the control group,and the rest of the organs were not significant(P >0.05).The growth of RKBL group was stable,without special histopathology,and there was no significant difference in weight,hematology and biochemistry test compared with the control group(P > 0.05).4,mice injected with RKBL pregnancy its palaces STAT3 expression quantity was suppressed,thus infer red kidney beanplant lectins in pregnancy is a possible way to inhibit the activation of STAT3 and the expression of STAT3,thus achieve the result of the termination of pregnancy.
