Objectives To investigate the expression and significance of STOML2 in renal interstitial fibrosis rats caused by unilateral ureteral obstruction(UUO);and the relationship with the markers of epithelial-mesenchymal transition(E-cadherin,Vimentin).To provide the experimental basis for clinical prevention on renal tubulo-interstitial fibrosis.Methods Thirty-six SD rats were divided into sham-operation group(n=18)and UUO model group(n=18).Rats were sacrificed 3,7,14 day after operation established for model group(n=6 for each day),and the renal tissues of the operated side were obtained from HE staining,Van Gieson staining.Immunohistochemical staining were used to evaluate the degree of renal tubulointerstitial fibrosis,collagen deposition and detect the expression of STOML2,as well as EMT-related markers E-cadherin,Vimentin in renal tissues.Correlation analysis was carried out to confirm the relationship among STOML2,E-cadherin and Vimentin.Results 1 HE staining: Renal tissues were not significant changes in sham group,occasionally accompanying with interstitial infiltration of inflammatory cells.In model group,renal interstitial lesions broadened with obvious infiltration of inflammatory cells,Epithelial cell vacuolar degeneration and renal tubular ectasia.With the prolongation of obstruction time,the degree of renal interstitial fibrosis was increased,which indicated that the model of renal interstitial fibrosis was successful.2 Semi-quantitative analysis of Van Gieson: The difference of collagen deposition score between sham group and the model group was statistically significant(P<0.5).The scores of model group 3days,7days,and 14 days were individually as(1.40±0.127),(2.40±0.179),(3.27±0.103),which was just to indicate that with the aggravation of obstruction,the deposition of collagen gradually increased.3 The semi-quantitative analysis of immune-histochemical staining:(1)Expression of STOML2 in renal tissues of rats the expression of the two groups was statistically significant(P<0.5).In sham group,only a small amount of STOML2 expressed in the distal renal tubules widened to the top,while in model group the renal interstitial expressed STOML2 protein.The scores in model group of 3days,7days,and 14 days were individually as(3.28±0.371),(4.38±0.286),(5.48±0.204).In other words,with the obstruction degree increasing,the expression level of STOML2 increased gradually.(2)Expression of E-cadhrin,Vimentin in renal tissues of rats The difference between sham group and model group was statistically significant(P<0.5).The expressions of E-cadherin in model group of 3days,7days,14 days were individually as(4.57±0.294),(3.10±0.518),(1.55±0.309);the expressions of Vimentin in model group of 3days,7days,14 days were individually as(2.93±0.516),(4.32±0.371),(5.42±0.286).These were to say epithelial-mesenchymal transition of renal tubular epithelial cells had happened,along with Renal interstitial fibrosis.4.Correlation analysis In model group,the expressions of STOML2,E-cadherin and Vimentin had strong relation.Conclusions 1 In UUO animal model,the expression of STOML2 was gradually increasing with aggravation of fibrosis,which suggested that STOML2 may be very vital in the process of renal fibrosis.2 The relationship between the expression of STOML2 protein and renal tubular epithelial mesenchymal transition marker proteins(E-cadherin,Vimentin)were very intimate,indicating that STOML2 could involve in the process of renal interstitial fibrosis...
