Objective:Influenza is a kind of infectious disease that is easy to spread or infect,and often causes pandemic.Because of the variability of influenza virus,existing vaccines are often ineffective against new epidemic strains,and some antiviral drugs also show resistance tendency,so it is imperative to explore new effective treatments.Some studies have shown that the IGF1 gene may play an important role in influenza infection,in this study,we filtrated and verified the micro RNAs that have a targeted relationship with IGF1 gene,and studied its role in influenza infection.This provides a new idea for studying the pathological mechanism of influenza and developing influenza treatment methods.Methods:1.Use the micro RNA target gene prediction database to search for micro RNAs that may have a targeted relationship with the IGF1 gene;2.Using chicken embryos to amplify PR8 influenza virus and constructing a cell model of PR8 influenza virus infecting A549 cells;3.Real-time fluorescence quantitative PCR was used to test the expression of selected micro RNAs in PR8 influenza-infected A549 cells;4.The dual luciferase reporter gene system was used to confirm the targeted relationship between the selected micro RNAs and IGF1;5.Observe the effect of these micro RNAs targeting the IGF1 in influenza infection.Results:1.Overlapping results from each database,selecting miR-1-3p,miR-29a-3p,miR-29b-3p,miR-29c-3p,miR-206-3p,miR-129-5p,miR-19a-3p,miR-19b-3p,miR-199a-3p,miR-362-3p,miR-30d-5p were the targets of the study;2.Some miRNA are down-regulated in A549 cells following influenza infection:miR-1-3p,miR-29a-3p,miR-29b-3p,miR-29c-3p,miR-19a-3p,miR-19b-3p,miR-362-3p;3.Mi RNAs that have a targeted relationship with the IGF1 gene include: miR-19a-3p,miR-29a-3p,and miR-29c-3p;4.The over-expressed miRNA decreases the expression of inflammatory cytokines IL-8 and influenza virus M1 genes that increase expression in influenza infection,and suppresses the expression of miRNA to increase it.Conclusion:1.Mi R-19a-3p,miR-29a-3p and miR-29c-3p may have a targeted relationship with IGF1 gene;2.The micro RNAs what target to IGF1 gene can regulate influenza virus infection at the cellular level.
