Objectives To observe the effect of Tiaogeng Jie Yu Fang on behavior and sex hormones in MD rats,and to explore the mechanism of Tiaogeng Jie Yu Fang on MD by regulating the downstream target of BDNF and activating P-P38 MAPK pathway and ERK1/2 pathway in rat hippocampus.Methods 72 healthy females were selected from 3 to 4 months,and they were randomly divided into sham operation group,model group,and Tiaogeng jieyu fang low,medium,high dose group,and control group of western medicine group,in addition to the sham operation group,with bilateral oophorectomy,chronic unpredictable stress(CUMS)and the manufacturing method of suppression climacteric depression model.Were divided into sham operation group,model group,tiaogeng Jieyufang low,medium and high dose group,western medicine group(Dai Lixin),respectively in the model for 0 days,7 days,14 days,21 days,,before gavage for 7 days,14 days,21 days for determination of body weight of rats,open the experimental box,sugar test,forced swimming test behavioral indicators,were sacrificed after 21 days of treatment,the content of serum was measured by ELISA method,FSH,LH E2,hippocampus HE staining was used to observe the morphological changes,immunohistochemical detection of BDNF protein in hippocampus and expression of Western-blot in hippocampus of P-P38 MAPK detection,RT-PCR detection ERK1/2 the level of the hippocampus.The data were analyzed by SPSS17.0 software.The experimental results were expressed with mean mean standard deviation(x + s).Single factor analysis of variance was used in multiple groups to satisfy normal and homogeneous variance.22 was compared by LSD test.When the variance was not homogeneous,Dunnett 's T3 method was used to test.The difference was statistically significant in P<0.05.Results 1Compared with the model group,rats in the high-dose group had a significant increase in body weight,significantly increased open-cell test scores,increased sugar consumption,and decreased immobility time of forced swimming(P<0.05).2Compared with the sham-operated group,serum E2 in the model group decreased,and FSH and LH increased in the model group(P<0.05).Compared with the model group,E2 in the high,medium,high dose group and western medicine group increased,and FSH and LH decreased.(P<0.05).3 The neurons in the hippocampal CA1 and CA3 regions of the model group were irregularly arranged and dispersed,and many glial cells were seen.In the treatment group,the neurons in the CA1 and CA3 regions were arranged in a regular and tight manner,with a small amount scattered in glial cells.4 The expression of BDNF in CA1 and CA3 regions of hippocampus in sham-operated rats was more than normal.The morphology of cells was intact and densely packed.The number of BDNF-positive cells in CA1 and CA3 of hippocampus in the model group was less,and the arrangement was sparse.The vacuoles and wrinkles were observed.Shrinking,the number of positive cells in the treatment group increased to varying degrees.5 Compared with the sham-operated group,the gray value of P-P38 MAPK in the hippocampus of the model group was increased(P<0.05),and the gray value of each group of Xiyu Yuyu and Western medicine was lower than that of the model group(P<0.05).In group comparison,the gray value of the high-dose Chinese medicine group decreased(P<0.05),and there was no statistical significance compared with the other two traditional Chinese medicine groups(P>0.05).6 The ERK1/2m RNA reverse transcription level in the model group was significantly lower than that in the sham group.The ERK1/2m RNA level in the hippocampus of the treatment group was higher than that in the model group(P<0.05).The high dose group was significantly higher than the model group.Conclusions 1 The experimental results showed that the depression of MD model rats could be improved by the recipe of Tiaogeng jie yu fang.2 Tiaoyu Jieyu Decoction has better regulation of endocrine hormone levels in MD model rats.3 "Tiaogeng jie yu fang" increased the expression of ERK1/2 by increasing BDNF levels,increased the expression of ERK1/2,and reduced the content of P-P38 MAPK to exert its antidepressant effect.
