Objectives The aim of this study is to detect the expressions of Forkhead box M1(FOXM1)and A disintegrin and metalloproteinase 17(ADAM17)in human cervical squamous cell carcinoma(CSCC),and analyze the relationships between FOXM1,ADAM17 and clinical significance.Methods Clinical data and paraffin specimens were collected from a few of Hospital in CHENG DE city(January 2010-December 2017),including in 89 cases of CSCC(52 cases of nonkeratinizing type and 37 cases of keratinizing type),23 cases of low-grade squamous intraepithelial lesion,65 cases of high-grade squamous intraepithelial lesion,30 cases of normal cervical mucosa.FOXM1 and ADAM17 protein expression were detedcted in these tissues by immunohistochemistry staining or western blot.The correlationship between FOXM1 and clinic-pathological data and the one between ADAM17 and clinicpathological data were also analyzed respectively.Five cases of fresh tissue in low differentiation CSCC,high differentiation CSCC and cervical normal mucosa were collected from a few of Hospital in CHENG DE city l(June 2017-December 2017).Western blot assay the expressions in these fresh tissues.Results 1 Western blot results:(1)The optical density(OD)of FOXM1 incontrol,keratinizing CSCC group and nonkeratinizing CSCC group was 0.14±0.04,0.35±0.06 and 0.57±0.11 respectively.Compared with control,the expression levels of FOXM1 protein in keratinizing CSCC and nonkeratinizing CSCC were significantly increased(P<0.05).The expression of FOXM1 protein in nonkeratinizing CSCC was also significantly higher than that of keratinizing CSCC(P<0.05).(2)The OD of ADAM17 in control,keratinizing CSCC group and nonkeratinizing CSCC group was 0.87±0.13,1.22±0.079 and 1.58±0.16 respectively.Compared withcontrol,the expression levels of ADAM17 protein in keratinizing CSCC and nonkeratinizing CSCC were significantly increased(P<0.05).The expression of ADAM17 protein in nonkeratinizing CSCC was also significantly higher than that of keratinizing CSCC(P<0.05).2 Immunohistochemical results:(1)FOXM1 protein was expressed in nuclei with yellow or brown color.The expression index of FOXM1 in control,low-grade squamous intraepithelial lesion,highgrade squamous intraepithelial lesion groups,keratinizing CSCC group and nonkeratinizing CSCC group was 0.34±0.09,1.31±0.18,2.98±0.22,4.41±0.29 and 7.24±0.28 respectively.The FOXM1 expression index of nonkeratinizing CSCC was significantly higher than that of control,CIN groups and keratinizing CSCC(P<0.05).The expression index of keratinizing CSCC was significantly higher than that of control and CIN groups(P<0.05).Compared with control,FOXM1 expression in CIN groups were higher with increased grades(P<0.05).(2)ADAM17 protein was expressed in cytoplasm with yellow or brown color.The expression index of FOXM1 in control,low-grade squamous intraepithelial lesion,high-grade squamous intraepithelial lesion groups,keratinizing CSCC group and nonkeratinizing CSCC group was 0.17±0.07,0.38±0.09,2.03±0.18,4.72±0.30 and 7.79±0.30 respectively.The ADAM17 expression index of nonkeratinizing CSCC was significantly higher than that of control,CIN groups and keratinizing CSCC(P<0.05).The expression index of keratinizing CSCC was significantly higher than that of control and CIN groups(P<0.05).Compared with control,ADAM17 expression in CIN groups were higher with increased grades(P<0.05).(3)The overexpression of FOXM1 and ADAM17 was associated with the degree of differentiation,but there was not related with patient age,FIGO stage(data only from stage I and stage IIA)and lymph node metastasis of CSCC.(4)The expression of FOXM1 and ADAM17 in the squamous cell carcinoma of the cervix was significantly positive correlated(r=0.664,P<0.05).Conclusions The expression of FOXM1 and ADAM17 are increased in CSCC,showing a positively correlationship.Overexpressions of FOXM1 and ADAM17 in CSCC are positively associated with the degree of differentiation,although there are no effect on patients age,FIGO stage and lymph node metastasis.
