Proanthocyanidin is a kind of polyphenol mixture with strong antioxidant activity,which is widely stored in the food resources such as dragon fruit,grape seed,acanthus fruit and blueberry.Procyanidins can react with free radicals and metal ion to product them a strong anti-aging effect because of phenolic hydroxyl.This paper studies the effect of procyanidin B2 anti-aging on metabolites of plasma and gut microbiota based on the LC-Q/TOF-MSn and Illumina high-throughout sequencing technology.The aim of research is to explore the mechanism of anti-aging,which provides a theoretical basis for the development and utilization of procyanidins in the future.The results were as follows:?1?Results showed that the procyanidin B2 significantly improved activity of T-AOC,SOD,GSH-Px of liver,brain,colon,and duodenal in the aged mice?P<0.05?.The level of MDA was decreased significantly in the aged mice treated with procyanidin B2.The open-field test implied that the procyanidins B2 significantly increased the number and time to the central region of aged mice,and increased the total distance of aged mice.In Morris water maze,the number of crossings,time spent,and distance in the target quadrant were significantly increased in the mice treated with the procyanidin B2,and the time of first platform was reduced,compared with aging group.These indicates that procyanidin B2 can improve the learning and memory ability of aged mice,increased the anti-oxidative ability of tissues,which in order to delay the process of aging.?2?The metabolites of plasma were obtained by LC-Q/TOF technology.We found that the level of LysoPE,PC,oleic acid,linoleic acid,carnitine,pantothenic acid,creatine,and taurocholic acid were significantly increased in the mice treated with procyanidin B2.Moreover,the level of pyruvic acid,D?-?-?-hydroxybutyric acid,hippuric acid,2-Methyl hippuric acid,taurorsodexycholic acid,cholic acid,7-ketodeoxycholic acid were significantly decreased.We found that procyanidin B2regulate such as glycerophospholipid metabolism,biosynthesis of unsaturated fatty acids,fatty acid metabolism,saturated fatty acid metabolism,phenylalanine metabolism,taurine and hypotaurine metabolism,bile acid biosynthesis and metabolism and so on.These were showed that the anti-aging effect of B2 could be related a variety of metabolites and they closely related to each other and affect multiple metabolic pathways.Since aromatic compounds have been metabolized into benzoic acid in the intestinal microorganisms,they were combined with glycine in the liver and kidney to form the hippuric acid.Since abnormal secretion of bile acid caused a direct result of structure change of the intestinal flora,which implied the procyanidin B2 could change the structure of intestinal flora.?3?Through analysis of?11umina,212 OTU were obtained in the three groups,5 OTU were found between control group and the aging group,and 11 OTU were between aging group and the PB2 group.Expect that the Shannon value of PB2group was lower than the aging group?P<0.05?,there was no significant difference in other diversity indexes such as Ace.Procyanidin B2 can improve the B/F value,so that its value is closer to the control group?P<0.05?.The procyanidin B2 can regulate the level of Bacteroidetes,Roseburia,Lachnospiraceae in order to increase the content of short chain fatty acid?P<0.05?,which suggested that the procyanidin B2 could adjust the structure of intestinal flora and inhibit the disturbance of the intestinal flora caused by aging.In a word,procyanidin B2 possesses a strong ability of antioxidant,and could significantly improve the dynamic balance of oxidation and reduction in aged mice.Procyanidin B2 could regulate TCA,taurine,glycerophospholipid metabolism and other metabolic pathways to adjust overall system metabolism profile of body and repaired the construction and balance of intestinal flora casued by aging.Procyanidin B2 can play an important role of anti-aging to slow down the process of aging,in order to improve quality of elderly.
