Red Ginseng is a traditional Chinese herbal medicine,made of white ginseng cooked and processed.It has been reported that the polysaccharide has many activities,such as anti-tumor and antioxidant.Its active components are mainly saponins and polysaccharides.There are few reports on polysaccharides of Red Ginseng,especially on the structure and structure-activity relationship of its polysaccharides.In order to understand the composition and structure of polysaccharides from Red Ginseng,we designed the research contents of this subject.The specific results are as follows:In this paper,Red Ginseng polysaccharide provided by Jilin Ji’an Yisheng Pharmaceutical Co.,Ltd.was used as experimental material,the polysaccharide was precipitated with 60%ethanol,collecting sediments after centrifugation,after drying,the water-soluble polysaccharide from Red Ginseng(WRGP)was obtained.The total sugar content was 80.1%,uronic acid was 23.9%,the protein content was 2.2%,the salt content was12.4%.The monosaccharide composition analysis showed that the WRGP was mainly composed of Glc(53.8%)and GalA(27.9%),In addition,it also contains Gal(9.5%)and Ara(5.4%),and a small amount of Rha(1.6%)and Man(0.7%).DEAE-Cellulose ion exchange chromatography is used to classify WRGP,The neutral fraction(WRGPN)was obtained by distilled water elution and the acidic fraction(WRGPA)eluted by 0.5M NaCl.WRGPA was further purificated and obtained WRGPA-N,WRGPA-1,WRGPA-2,WRGPA-3 and WRGPA-4.WRGPA-1WRGPA-4 were purificated by using gel chromatography,finally obtained homogeneous fractions WRGPA-1a,WRGPA-2a,WRGPA-2b,WRGPA-3b,WRGPA-3c,WRGPA-4b and WRGPA-4c.Monosaccharide composition,enzymolysiss,methylation,FT-IR and 13C NMR analysis showed that WRGPA-1a and WRGPA-2a were mainly composed of RG-I,AG-I was attached to the main chain as a side chain;WRGPA-2b,WRGPA-3b,WRGPA-3c,WRGPA-4b and WRGPA-4c were mainly composed of GalA(56.9%77.3%),they were typical HG pectin,composed of a backbone of a linear-α-1,4-linked D-galacturonic acid with different degrees of methyl-esterification.A small number of RG-I structures are linked to the main chain of HG.
