Objective:The global incidence of melanoma is high,and the prevalence and mortality are high.However,its specific pathogenesis has not been elucidated.Traditional therapy,surgical treatment,radiotherapy,chemotherapy and the latest molecular targeting therapy can not achieve satisfactory results.Recent studies have shown that classical artemisinin-derived dihydroartemisinin,a first-line antimalarial drug,has extensive antitumor activity.The aim of this study is to investigate the effect of Dihydroartemisinin on proliferation of melanoma cells and its underlying molecular mechanism,so as to provide reliable experimental evidence for the clinical application of dihydroartemisinin in the treatment of melanoma.Methods:The effects of dihydroartemisinin on the proliferation of two cell lines were detected by MTS method.The effects of dihydroartemisinin on cell colony formation were detected by the tablet cloning technology.Finally we used the flow cytometry to detect the cell cycle distribution after treatment with dihydroartemisinin,and used the western blot to detect the changes of cycle-related proteins.Results:1.Different concentrations of dihydroartemisinin can inhibit the proliferation of melanoma cells A2058 and G361,and show concentration-and time-dependent effects.2.Different concentrations of dihydroartemisinin can inhibit the colony formation of melanoma cells A2058 and G361.3.Different concentrations of dihydroartemisinin can cause S phase and G2/M phase block in melanoma cell A2058,and G0/G1 phase block in melanoma cell G361.Conclusions:Dihydroartemisinin can inhibit cell proliferation by causing the corresponding cycle arrest of melanoma cells A2058 and G361.
