Background: Diabetes mellitus is a metabolic disease characterized by high blood sugar.It is the world's third-largest chronic disease threatening human health.Diabetic nephropathy is the most important cause of death in patients with diabetes.The pathogenesis of diabetic nephropathy is complex,has not been fully elucidated.Autophagy is very important to maintain the intracellular organelles function and nutrition environment.Autophagy also participates in the metabolism of systemic balance and its disorders can cause or accelerate the development of metabolic disorders.Studying found that the anomaly of autophagy participating in the occurrence and development of diabetic nephropathy.Exercise can enhance immunity,improve the vivo environment,adjust the sugar,fat,and endocrine metabolism,improve cardiopulmonary function,etc.It has been found that exercise can activate autophagy degradation organelles.So the reduce of autophagy may be one of the pathogenesis of diabetic nephropathy,the improvement of autophagy may become the new target for clinical treatment of diabetic nephropathy.Objective: To explore whether the moderate exercise can enhance autophagy to protect diabetes of renal injury in mice.?Methods: 50 mice adaptive points cage for a week.Grouping,20 mice were randomly selected as control group,fed chow,the remaining 30 feed feeding high fat and sugar.Average daily record each rat eating,drinking,and say once a week.After six weeks,mice fed a high fat and sugar feed,fast water,12 h,according to the dose of 100 mg/kg body weight chain of intraperitoneal injection of 1% of urea with cephalosporins type(STZ)induced citric acid ?buffer solution.Injection of STZ mice were determined after ten days of fasting plasma glucose greater than 11.1 mmol/L as a basis of ?diabetes model.The swimming mice were randomly divided into control group and normal group,Cheng Mo swimming mice were randomly divided into model group and diabetes group,metformin group.Swimming,lavage metformin intervention treatment to be put to death in mice after seven weeks,pick the eyeball blood,renal function tests,serum samples from organization,on the kidney tissue of HE staining,immunohistochemistry and Western blot detection related proteins.Results: A significant rise in model group mice serum insulin levels,and diabetes swimming group serum insulin level has decreased in the model group.Biochemical indicators of renal function in model mice also showed increased obviously,the kidney weight/body weight ratio also increased significantly,and compared with model group,diabetes group swimming reduced the renal function indexes,kidney weight/body weight ratio,too,fell,illustrate swimming exercise on diabetes mice kidney damage.WB results show that: compared with control group,the expression of LC3 in model group decreased significantly(P < 0.05),and compared with model group,diabetes swimming group LC3 increased(P < 0.05).compared with the control group,model group P62 expression increased significantly,and compared with the model group,diabetes swimming group p62 have decreased(P < 0.05).Immunohistochemical results show that: compared with model group,diabetes swimming group LC3 expression increased,the expression of p62 fell.Results show that the swimming movement enhanced the level of autophagy.Conclusion: Moderate exercise can enhance autophagy to protect ? type of renal injury in mice.
