Objective: To investigate the effect of silencing Mig-7 gene on biological characteristics of prostate cancer PC3 cell,including invasion? migration,and the forming of vasculogenic mimicry.Methods:Human prostate cancer cell line PC3 was cultured and siRNA was used to silence Mig-7 gene in prostate cancer PC3 cells.The expression of Mig-7 mRNA was detected by fluorescence quantitative real-time PCR,the change of Mig-7 protein was detected by Western blot,and the most effective interference group were selected.The experiments were divided into PC3 group,PC3+NC siRNA group and PC3+Mig-7 siRNA group,after transfection of 48 h,three groups of cells were cultured in vitro,and the number of mimicry was observed under inverted microscope.At the same time,Transwell method and cell scratch test were used to observe the invasion and migration ability of three groups.Results:The results of Real-time fluorescence quantitative PCR and Western-blot showed that siRNA silenced Mig-7 gene in prostate cancer PC3 cells,and the mRNA and protein expression levels of PC3 cells were significantly decreased,and the Mig-7 gene silencing efficiency was over 60%.The results of Transwell and scratch test showed that silencing Mig-7 gene,the invasion and migration ability of PC3 cell decreased;The results of the three-dimensional culture showed that the number of angiogenic mimicry of Mig-7 gene and prostate cancer PC3 cells was significantly reduced.Conclusion:Silencing Mig-7 gene can decrease its invasion and migration ability,and the forming of vasculogenic mimicry.
