Background As a systemic inflammatory response syndrome,sepsis can lead to multiple organ dysfunction,the leading cause of death in critically ill patients.Sepsis is the most important complication of sepsis,and the incidence of sepsis is as high as 40%.It is found that mitochondrial dysfunction is important in the development of sepsis and is an important field in the study of sepsis.Encoded by mitochondrial DNA specific expression of ND1,Cytb,Atp6 three gene expression level to a certain extent,can reflect the function of mitochondria,if mitochondrial function is impaired,ND1,Cytb,Atp6 gene expression level will be lowered.And a large number of studies have shown that CUR can influence different levels of the expression of inflammatory mediators,which play a role of immune regulation,anti-inflammatory and anti-cancer,the sepsis sexual protection of myocardial injury.Objective To observation of mt ND1,mt Cytb,mt Atp6(CLP)in the cecum ligation perforation technique method to establish the rat myocardial injury and sepsis after using curcumin intervention level of dynamic change,explore the curcumin for mitochondrial mt ND1 sepsis myocardial injury,the influence of mt Cytb,mt Atp6 gene expression.Methods The 30 SD rats were randomly divided into six groups: Sham operation 6 hours group,24 hours group,model(CLP)6 hours group,24 hours group,intervention(CUR)6 hours group and 24 hours group,each group of taken five rats.The experimental model of classical sepsis in rats was performed by CLP.The sham operation group only were operated without ligation and perforation.CUR intervention group were intraperitonealled of 100 mg/kg CUR with Dimethyl phosphite alum configuration,concentration of 50 mg/ml to model,immediately after surgery.To observe the general conditions of the rats in 48 hours after the modeling.The blood samples were collected At corresponding time points,collected the rats blood samples to detected the levels of CK and CK-MB,gross morphology of the heart,HE stained of the pathological condition of myocardial tissue,used Real-time RT-PCR to test the expression of mt ND1,mt Cytb and mt Atp6 of the myocardial tissues.Results(1)Compared with Sham 6h group,the serum CK levels(975.20±134.83 vs 1378±136.10,p <0.01),CK-MB levels(282.20±17.67 vs 368.40±25.49,p <0.01)in rats of Sham 24 h group decreased significantly,the serum CK levels(2877.20±307.32 vs 1378 ±136.10,p<0.01),CK-MB levels(817.20±19.98 vs 368.40±25.49,p <0.01)in rats of CLP 6h group increased significantly;Compared with the CLP 6h group,the serum CK levels(1839.00±171.13 vs 2877.20±307.32,p <0.01),CK-MB levels(461.60± 43.04 vs 817.20±19.98,p <0.01)in rats of CLP 24 h group,and the serum CK levels(2474.00±102.24 vs 2877.20±307.32,p <0.05),CK-MB levels(656.40±55.35 vs 817.20±19.98,p <0.01)in rats of CUR 6h group decreased significantly;Compared with the CLP 24 h group,the serum CK levels(975.20 ±134.83 vs 1839.00 ±171.13,p <0.01),the level of CK-MB(282.20± 17.67 vs 461.60±43.04,p <0.01)in rats of Sham 24 h group and the serum CK levels(1365.40±140.40 vs1839.00±171.13,p <0.01),the level of CK-MB(361.80±58.26 vs 461.60±43.04,p <0.01)in rats of CUR 24 h group decreased significantly;Compared with the intervention 6h group,the serum CK levels(1365.40±140.40 vs 2474.00±102.24,p < 0.01),CK-MB levels(361.80±58.26 vs 656.40±55.35,p <0.01)in CUR 24 h group significantly decreased.(2)Pathologic changes of myocardial tissue in rats: no obvious pathological changes were observed in the myocardial tissues of the rats in 6h group and 24 h group.In the model 6h group,a large number of myocardial cell edema and vacuolar formation were seen in the rats,and the lesion was characterized by focal granular degeneration.The intercellular mass was increased and the exudation was obvious,and there were a large number of inflammatory cells infiltrating around.In the 24 h group,the hyperemia and vacuolar formation of myocardial cells in rats were significantly reduced,and the infiltration of inflammatory cells decreased,and the regenerated cardiomyocytes were seen.Curcumin(CUR)interfered in the myocardial cell lesion of 6h group,and the edema was reduced and a small amount of necrosis was observed.The intervention of curcumin(CUR)in the 24 h group showed significantly decreased myocardial edema and decreased inflammatory cell infiltration.Myocardial histopathological scores: There was no significant difference in myocardial pathological score between the sham-operated 6h group and the sham-operated 24 h group,The difference was not statistically significant.High(p <0.01);Compared with the 6h group,pathological scores(p <0.01)in the 24 h group and pathological scores(p < 0.05)in the 6h group were significantly higher;24h group compared with the 24 h group.The scores(p <0.01)and pathological scores(p <0.05)decreased significantly in the 24-hour intervention group,and the intervention 24-hour group(p <0.01)significantly decreased compared with the 6-hour intervention group.(3)mtND1,mt Cytb,and mt Atp6 expression levels in Rat myocardial tissue : Compared with the Sham 6h group,the myocardial tissue mt ND1 levels(1.53±0.6 vs 1.00±0.01),mt Cytb(2.08± 0.0 vs 1.00±0.07,p <0.05),mt Atp6 levels(0.30±0.2 vs 1.00±0.27)in the Sham 24 h group,and mt ND1(2.30±1.6 vs 1.00±0.01),mt Cytb(0.67±0.48 vs 1.00±0.07),mt Atp6 levels(0.35±0.2 vs 1.00±0.27,p <0.01)in the CLP 6h group;Compared with the CLP 6h group,the levels of mt ND1(2.30±2.17 vs 2.30±1.66),mt Cytb(0.43±0.46 vs 0.67±0.48),and mt Atp6(0.30±0.21 vs 0.35±0.22)in the 24 h group,and mt ND1(1.22±0.27 vs 2.30±1.66),mt Cytb(0.56±0.14 vs 0.67 ±0.48),mt Atp6(0.63±0.21 vs 0.35±0.22)in the CUR 6h group were decreased,but without statistically significant.Compared with the CUR 6h group,the mt ND1 levels(0.80±0.38 vs 1.22±0.27),mt Cytb(0.13±0.09 vs 0.56±0.14,p <0.01),mt Atp6 levels(0.35±0.09 vs 0.63±0.21,p <0.05)in CUR 24 h group decreased significantly.Compared with the CLP 24 h group,the mt ND1 levels(1.53±0.63 vs 2.30±2.17),mt Cytb(2.08±0.00 vs0.43±0.46),mt Atp6 levels(0.30±0.22 vs 0.30±0.21)in Sham 24 h group and the levels of mt ND1(0.80±0.38 vs 2.30±2.17),mt Cytb(0.13±0.09 vs0.43±0.46),mt Atp6 levels(0.35±0.09 vs 0.30±0.21)in the CUR 24 h group were changed,but without statistically significant.Conclusion CUR has an obvious protective effect on myocardial injury in sepsis rats,possibly by regulating the expression level of mt ND1,mt Cytb and mt Atp6,and protecting the mitochondrial function of myocardial tissue.
