Objective The recovery of nerve injury has become a key problem to be solved in clinical practice.It is known that PTEN gene is a tumor suppressor gene.Some experiments have proved that PTEN gene can inhibit peripheral motor neuron axon regeneration.Inhibition or knocking out PTEN gene contributes to peripheral motor neuron axon regeneration.Therefore we intend to confirm the effect of PTEN gene on axon regeneration by detecting the expression of PTEN gene after sciatic nerve injury and the regeneration of peripheral sensory neurons after inhibiting PTEN gene expression.Method 1.In this study,adult female ICR mice(institute of cancer research)sciatic nerve transection model(Axotomy)was used to extract DRG of normal adult female ICR mice and adult female ICR mice 3 days after sciatic nerve transection.The expression of PTEN gene after sciatic nerve injury was detected by Western-Blot,RT-PCR and immunohistochemical staining.2.DRG of normal adult female ICR mice and axotomy mice were cultured,and different concentrations of PTEN inhibitor were added into each group.After 24 hours of culture,the axon length was measured,and the effect of inhibiting PTEN gene expression on axon regeneration of peripheral sensory neurons of normal female ICR mice and adult female ICR mice after sciatic nerve transection was detected.3.DRG of normal adult female ICR mice was divided into two groups for cell culture.The control group was added with different concentrations of PTEN inhibitors,the experimental group was added with different concentrations of PTEN inhibitors,and m TOR specific inhibitor rapamycin was added.After 1 day of cell culture,the length of axon was measured,and whether the addition of PTEN inhibitors and rapamycin would affect the growth of axon was observed.Result After sciatic nerve injury,the expression of PTEN gene was detected by Western-Blot and RT-PCR.The expression of PTEN gene was significantly decreased.After the expression of PTEN gene was inhibited,the cell culture results of normal adult female ICR mice and axotomy treated adult female ICR mice showed that the axon length was significantly longer after adding PTEN inhibitor than that without adding PTEN inhibitors,so inhibiting the expression of PTEN gene could promote the axon growth of sensory neurons.The axon length of the experimental group with PTEN inhibitors and rapamycin did not change significantly compared with the control group with PTEN inhibitors alone.Conclusion Axon regeneration after peripheral sensory nerve injury is a complex biological process which can be strictly controlled by different genes.PTEN gene could regulate the axon regeneration of peripheral sensory neurons.Inhibition of PTEN gene expression can promote axon regeneration of peripheral sensory neurons.
