Due to the decreased ability of muscles to produce strength or weakened nerve activity,fatigue is a comprehensive physiological phenomenon.Muscle fatigue has two main reasons caused by.The ability of the nerve to produce a continuous signal?nervous fatigue?is limited.The ability to contract the muscle fibers is diminished?metabolic fatigue?.Alcoholic liver disease?ALD?is a complicated systemic and multisystemic disease.Alcoholism is a major cause,leading to increased alcoholic liver disease?ALD?global morbidity and mortality.On a global scale,the burden caused by alcohol-related diseases can not be underestimated.Ginseng,honeysuckle,Huang Jing,and dandelion are all traditional Chinese herbal medicines.In particular,ginseng is extremely valuable.This is well known and widely acknowledged during the medical care field.Lao Gu Niang Xing Gan No.1Drink is made using these four components,which having a natural,safe,non-toxic side effects etc.And also has a long history of medicine and food homology use.Objective:To study the effect of Lao Gu Niang Xing Gan No.1 Drink in anti-fatigue and acute alcoholic liver injury protection.Further elaborates its mechanism.Methods:In the study of the anti-fatigue activity of Lao Gu Niang Xing Gan No.1Drink,28 mice were divided into blank control group?CG?,positive control group?PG?,drug high-dose group?HDG?and low-dose group?LDG?.Through the weight-burdenning swimming experiment,the anti-fatigue test was carried out to determine the content of creatine kinase?CK?,lactic acid?LD?,urea nitrogen?BUN?and blood ammonia?PA?in serum.The levels of malondialdehyde?MDA?,superoxide dismutase?SOD?,liver glycogen?LG?levels and gastrocnemius muscles glycogen?MG?were detected in liver tissues.In the research of Lao Gu Niang Xing Gan No.1 in the experimental study of acute alcoholic liver injury.45 mice were divided into blank control group?BG?,positive control group?MDG?,alcohol model control group?AMG?,drink high dose group?HDG?and low dose group?LDG?.The experiment of acute alcoholic liver injury was carried out.The tolerance time and waking time of the mice were tested by using the positive reflection test.Alkaline phosphatase?AKP?,Glutamic Oxalacetic Transaminase?GOT?,Glutamic Pyruvic Transaminase?GPT?,Total Bilirubin?TBIL?and Triglyceride?TG?in serum were detected.The level of malondialdehyde?MDA?and triglyceride?TG?were detected in liver tissue.In the study of the mechanism of the protective effect of Lao Gu Niang Xing Gan No.1 on acute alcoholic liver injury,we further studied its participation in alcohol metabolism through liver tissue in mice with acute alcoholic liver injury to explore inflammation and fibrosis and fat metabolism and apoptosis related genes,by real-time PCR detection of ADH2,ALDH2,TGF-?1,?-catenin,PPAR-?gene mRNA expression,and by Western Blot detection of PPAR-?,AMPK,Akt protein expression,To further study the molecular mechanism of the protection of"Lao Gu Niang Xing Gan No.1",which against acute alcoholic liver injury from molecular level.Results:According to the anti-fatigue test results,the Lao Gu Niang Xing Gan No.1can significantly improve swimming time.The positive control group,high-dose and low-dose groups had statistically significant difference compared with the control group?P<0.05?.Muscle glycogen,hepatic glycogen and super oxide dismutase?SOD?levels increased.The level of hepatic glycogen?LG?and Muscle glycogen in the high-dose group of drink groups was significantly higher than that in the control group?P<0.05?.The levels of superoxide dismutase?SOD?of high and low doses of beverages groups were higher than those of the control group?P<0.05?.Lao Gu Niang Xing Gan No.1 can reduce the level of creatine kinase?CK?,lactic acid?LD?,blood urea nitrogen?BUN?,ammonia,malondialdehyde?MDA?.There was a statistically difference between the high and low beverage dose groups with the control group?P<0.05?;In the acute alcohol poisoning experiment,the intoxication incubation period of the drink group was increased,and the sleeping time was shortened,and all the indicators were significantly reduced.The levels of creatine kinase?CK?,lactic acid?LD?,urea nitrogen?BUN?,blood ammonia and malondialdehyde?MDA?were reduced.In the study of the effects of acute alcoholic liver injury in mice,compared with the alcohol model group,beverages adminstered mice increased the alcohol tolerance time,the period from wakefulness to lethargy is prolonged,the period from lethargy to wakefulness is shortened,and both have statistical significance.The serum levels of AKP,GOT,GPT,TG,and TBIL in the serum of the high-and low-dose group were significantly detected lower than those in the alcohol model group,and the levels were closed to the MDG.All had statistical significance?P<0.05?.The contents of TG and MDA in the liver of high-dose and low-dose beverage groups were significantly lower than those in the alcohol model group,and all had statistical significance?P<0.05?.In the experiment to explore the protective mechanism of acute alcoholic liver injury in mice,the expression of ADH2,ALDH2,PPAR-?,AMPK and Akt genes in the liver of high-dose and low-dose beverage groups was significantly up-regulated compared with the alcohol model group,and down-regulate TGF-?1 and?-catenin expression,all results were statistically significant?P<0.05?.Conclusion:?1?The swimming test was carried out by weight burdenning in mice.According to the recorded swimming duration.In the high dose and low-dose group of Lao Gu Niang Xing Gan No.1 drink,the swimming exhaustion time was prolonged,and Lao Gu Niang Xing Gan No.1 drink had the ability to improve exercise endurance.?2?In the detection of serum creatine kinase?CK?,lactic acid?LD?,urea nitrogen?BUN?,and blood ammonia?PA?.Compared with the control group,the mice that were given the Lao Gu Niang Xing Gan No.1 drink were significantly different from those in the control group.It is possible that Lao Gu Niang Xing Gan No.1 drink has the effect of promoting lactic acid metabolism to reduce its accumulation,indicating that Lao Gu Niang Xing Gan No.1 drink has the effect of eliminating fatigue and improving the physical endurance of body.The high dose and low dose of Lao Gu Niang Xing Gan No.1 both can significantly reduce the accumulation of MDA in the serum mice level.But also increase the activity of SOD,reduce oxidative damage and inhibit oxidative stress,improve the body's antioxidant capacity.Give mice a high dose of Lao Gu Niang Xing Gan No.1 drink as 16ml/kg which can significantly increased the storage of glycogen and myoglycogen.Therefore,this study is aimed to explore the purpose of Lao Gu Niang Xing Gan No.1 drink for anti-fatigue function.It is proved that this kind of functional drink with ginseng and other main components can improve the ability of exercise,relieve fatigue and enhance the function of fatigue elimination.?3?During this experiment,using a one-time binge drinking as 53°alcohol,to establish the acute alcoholic liver injury model.The AKP,GOT,GPT,TG and TBIL in the high dose and low dose group of Lao Gu Niang Xing Gan No.1 were significantly lower than those in the alcohol model group.And they were basically close to the positive drug group level.The contents of TG and MDA in mice with Lao Gu Niang Xing Gan No.1 drink were significantly lower than those in the alcohol model group.The above results suggest Lao Gu Niang Xing Gan No.1 can enhance the body's antioxidant capacity to protect liver cells,and thus reduce the damage caused by alcohol.The function of metabolizing fat is tending back to normal level.?4?Compared with the alcohol model group,the tolerance time of the mice taking the Lao Gu Niang Xing Gan No.1drink high dose group and the low dose group was significantly increased.The sleeping time was significantly shorter than that of the alcohol model group,and the liver index was significantly decreased from that of the alcohol model group.?5?According to the pathological analysis.The alcohol leads to a disorderly arrangement of hepatic cords,dilated hepatocytes,blurred cell lines,and a large accumulation of lipid droplets in hepatocytes.However,there was No obvious swelling of hepatocytes in the subjects of Lao Gu Niang Xing Gan No.1 drink and low dose,and the inflammation caused by binge drinking decreased.There was no significant difference compared as the positive drug group.The results of this pathological sections were consistent with the results of the changes of various indicators during this experiment,reflects the Lao Gu Niang Xing Gan No.1 drink significantly alleviated the state of liver injury.And it had a good protective effect on the liver.?6?The initial mechanism of the protective effect of Lao Gu Niang Xing Gan No.1 on acute alcoholic liver injury in mice,is through up-regulating ADH2 and ALDH2 to promote ethanol metabolism,up-regulates AMPK to relieve inflammation,down-regulating TGF-?1and?-catenin to improving liver fibrosis status,and up-regulating the expression of PPAR-?to alleviate the dysfunction of lipid metabolism caused by alcohol,and then play a role in improving liver alcohol metabolism dysfunction caused by excessive drinking,liver inflammation and fibrosis status and lipid peroxidation metabolism.
