Objective:To analyze the association between single-nucleotide polymorphisms(SNPs)of-33 position at the Interleukin 4(IL-4)gene promoter region,-572position at the Interleukin 6(IL-6)gene promoter region and the susceptibility of Febrile seizures,then we can provide clinical guidance for early prevention and treatment.Methods:A case-control study was used in this research.66 cases of febrile seizures were taken as case groups,and 40 healthy children matched for the age of health examination at Bethune First Hospital of Jilin University as control.The patients in the case group met the diagnostic criteria for febrile seizures released in 2010by the International Association Against Epilepsy(ILAE).Blood samples ofchildren with febrile seizures and normal control group were collected.All blood samples were cryopreserved at-80?.Genomic DNA was extracted from blood samples by DNA extraction kit,and then geneotype and allele distribution was detected by polymerase chain reaction(PCR)and DNA sequencing technology.WPS tables were used to build data tables,and SPSS 18 software was appliedfor statistical analysis.Results:1.There are three genotypes at the-33 position of interleukin 4 gene promoter region in the FS group and control group,which were CC,CT and TT respectively.The genotype frequency was no significant difference in the FS group and the control group(X~2=4.624,P>0.05);the frequency of C allele was27.3%in the FS group and 15%in control group,T allele frequency was 72.7%in the FS group and 85%in control group.The allele frequency was significant difference between FS group and control group(OR:2.125;95%CI:1.031-4.381;P<0.05).2.There are three genotypes at the-572 position of the interleukin 6 gene promoter region in the FS group and control group,respectively CC,CG and GG.The genotype frequency was no significant difference between the FS group and control group(X~2=5.394,P>0.05);the frequency of C allele was 72%in FS group and 57.5%in control group,T allele frequency was 28%in FS group and42.5%in control group,the allele frequency was significant difference between FS group and control group(OR:1.898;95%CI:1.059-3.402;P<0.05).Conclusion:1.The polymorphism of the-33 positon at IL-4 gene promoter region may be associated with the pathogenesis of FS,the increase of C allele frequency may constitute risk factors for developing febrile seizures.2.The polymorphism of the-572 positon at IL-6 gene promoter region may be associated with the pathogenesis of FS,the increase of C allele frequency may constitute risk factors for developing febrile seizures.
