Objective:To investigate the correlation between the coronary atherosclerotic heart(CHD)disease and the expression level of miR-125a-5p and miR-193b in peripheral blood mononuclear cells of coronary atherosclerotic heart disease patients.Methods:we have collected 102 subjects in the study,and divided into four groups,including 25 cases of normal control group,22 cases of stable angina pectoris group,26 cases of unstable angina pectoris group and 29 cases of acute myocardial infarction group.Extracting subject's peripheral venous blood,then separating the mononuclear cells by the gradient density centrifugation method.The total RNA in mononuclear cells was extracted by TRIzol reagent and synthesis of cDNA by reverse transcriptio.The expression levels of miR-125a-5p and miR-193b in peripheral blood mononuclear cells were detected by real-time fluorescence quantitative PCR.The experimental data was analyzed by using SPSS 17.0 statistical software.Result:1.Compared with the normal control group,the expression level of miR-125a-5p was decreased significantly in the stable angina group(0.57 ± 0.56 vs.1 ± 0.55,P=0.002)?unstable angina pectoris group(0.37 ± 0.21 vs.1 ± 0.55,P=0.000)and the acute myocardial infarction group(0.36 ± 0.28 vs.1 ± 0.55,P=0.000).2.Compared with the stable angina group,the expression level of miR-125a-5p was decreased significantly in the unstable angina pectoris group(0.37 ± 0.21 vs.0.57 ±0.56,P=0.013)and the acute myocardial infarction(0.36 ± 0.28 vs.0.57 ± 0.56,P=0.007).3.There was no significant difference in the expression level of miR-125a-5p between unstable angina pectoris group and acute myocardial infarction group(0.37 ± 0.21 vs.0.36 ± 0.28,P=0.857).4.Compared with the normal control group,the expression level of miR-193b was decreased significantly in the stable angina group(0.53 ± 0.82 vs.1 ± 0.57,P=0.008),unstable angina pectoris group(0.26 ± 0.25 vs.1 ± 0.57,P=0.000)and the acute myocardial infarction group(0.25 ± 0.24 vs.1 ± 0.57,P=0.000).5.Compared with the stable angina group,the expression level of miR-193b was decreased significantly in the unstable angina pectoris group(0.26 ± 0.25 vs.0.53±0.82,P=0.003)and the acute myocardial infarction(0.25 ± 0.24 vs.0.53 ± 0.82,P=0.001).6.There was no significant difference in the expression of miR-193b between unstable angina pectoris group and acute myocardial infarction group(0.26± 0.25 vs.0.25 ±0.24,P=0.832).Conclusion:1.The expression level of miR-125a-5p was decreased significantly in peripheral blood mononuclear cells of coronary atherosclerotic heart disease patients,and the expression level was:acute coronary syndrome<stable angina pectoris<normal control.2.The expression level of miR-193b was decreased significantly in peripheral blood mononuclear cells of coronary atherosclerotic heart disease patients,and the expression level was:acute coronary syndrome<stable angina pectoris<normal control.
