Objectives:1.to analyze arsenic content in workers exposed to arsenic and evaluate arsenic load in workers exposed to arsenic.2.detect the expression of PANDAR gene and the influence of arsenic exposure workers on PANDAR methylation,and analyze the relationship between PANDAR gene expression and three kinds of urinary arsenic compounds.3.the effect of arsenic exposure on the expression of NEAT1 gene and its methylation,the changes in the expression of NEAT 1 gene by arsenic metabolites and the change of NEAT 1 gene expression after adding methyl donorMethod:1.the method of cluster sampling was used to extract arsenic exposed occupational workers in a arsenic plant as exposure group,and a control group was set up to collect urine samples and peripheral blood samples.2.HG-coldtrap-AAS method was used to determine the content of three arsenic compounds in iAs,MMA and DMA.The arsenic methylation index of TiAs,iAs%,MMA%,DMA%,PMI,SMI and so on were calculated.The effects of arsenic exposure on the expression of PANDAR gene in occupational workers;3.the genomic DNA of peripheral blood samples was extracted by centrifuge column method,and the methylation sensitivity high resolution dissolution curve was used to detect the effect of arsenic exposure on the methylation of PANDAR gene in occupational workers.4.real-time fluorescence quantitative detection of NEAT1 gene expression and methylation in cells exposed to different concentrations of arsenic.5.t test and non parametric test were used to analyze the effects of arsenic exposure on PANDAR expression,DNA damage and methylation,as well as arsenic and arsenic compounds in urine,and the effects of Spearman correlation analysis on the expression of PANDAR gene in urine,arsenic,arsenic and methylation.Result:1.Content of iAs,MMA,DMA and TiAs in urineThe median of urine concentrations of iAs,MMA,DMA and TiAs in exposed workers were 104.69(20.115645.03),156.43(20.59892.57)?g/ml,603.66(48.402172.25)?g/ml and 822.39(111.63-3526.1)?g/ml.The median of urine concentration of iAs,MMA,DMA and TiAs in the control group were 1.67(0.42,5.83)?g/ml,1.57(0.41,5.84)lg/ml,14.85(1.82,43.06)?g/ml and 17.98(2.95,54.28)?g/ml,under the urinary total arsenic biological contact limit.2.Expression of PANDAR gene in peripheral blood of peopleThe expression of PANDAR in the exposed group was significantly different from that in the control group(t =2.915,P<0.05),and the expression in the exposed group was significantly higher than that in the control group.There was a positive correlation between the expression of PANDA gene and iAs and MMA.There was a negative correlation between the expression of PANDAR gene and the rate of DNA methylation in peripheral blood,and the difference was statistically significant(R2=0.05,p=0.011.).3.the expression of NEAT 1 gene in A549 cells and MDA-MB-M231 cells under different arsenic concentrations.Compared with the control group,the expression of NEAT1 was different,and the value of P was<0.05,and the value of Pwas<0.05.Compared with the control group,the T values of the exposed groups were 4.177,4.229,4.730,and the MDA-MB-M231 cells were compared with the control group,and the T value of the exposed groups was 4.001,13.62,10.82),which could be considered as the arsenic concentration.The expression of NEAT1 gene increased.Under the combined action of SAM,the NEAT1 gene was expressed in A549 cells and MDA-MB-M231 cells,arsenic poisoning group and arsenic and SAM were combined,and compared with the control group,the expression of NEAT1 was different(t value in A549 cells,P value<0.05,t value in MDA-MB-M231 cells,<0.05)..As compared with arsenic and SAM,there were differences in NEAT1 expression(t value =3.494,P<0.05,=3.595 in MDA-MB-M231 cells in A549 cells,t in MDA-MB-M231 cells,P<0.05).The expression of NEAT1 gene expression was lower than that of single arsenic exposure group.Conclusion:1.arsenic and arsenic compounds in workers exposed to occupational arsenic exposure increased significantly,and arsenic exposure had potential occupational hazards to workers.The level of TiAs was significantly higher than that of the latest urinary total arsenic exposure limit established by the American government industrial hygienists conference,which is 35 g/ml.2.the experiment found that the expression of PANDAR gene in arsenic exposed workers increased,and the higher the exposure level of arsenic,the higher the expression of PANDAR gene,and the positive correlation between the expression of PANDAR gene and the iAS and MMA in the body urine.In 3.cell experiments,it was found that as the concentration of arsenic increased,the expression of NEAT 1 increased,and the arsenic metabolites did not increase the expression of NEAT 1.It was presumed that the metabolic process of arsenic resulted in the increase of gene expression.After adding SAM,the expression of NEAT 1 gene in the arsenic exposure group decreased.The inference is that SAM can accelerate the metabolism of arsenic in cells and counteract the effect of arsenite on NEAT1 gene expression.
