Objective:To study the expression of cannabinoid and cannabinoid receptor 2 in experimental rat type ? prostatitis and prostate tissue,and the influencing factors of expression and the accompanying pelvic pain changes.To investigate the relationship between cannabinoid and cannabinoid receptor 2 and chronic pelvic pain in experimental rat type ? prostatitis.Provide research ideas for the treatment of type? prostatitis pain.Methods:Twenty-five mature male SD rats were selected and divided into five groups(0.9%sodium chloride pelvic region subcutaneous and bilateral scapular subcutaneous and intraperitoneal injection),type ? prostatitis model/EAP group.(Complete Freund's adjuvant and prostate tissue mixed suspension pelvic region subcutaneous,bilateral scapular subcutaneous and intraperitoneal injection),after modeling,the prostate of 5 EAP rats were taken for HE staining detection modeling success rate;The remaining rats in the EAP group were further divided into three groups:5 intervention groups(?)(addition of cannabidiol was added to the EAP model group),and 5 intervention groups(?)(addition of cannabinoids based on the EAP model group.Agonist GW405833 intervention),intervention group(?)5(trampoma intervention based on EAP model group).After successful modeling,the intervention group(?)continuously intraperitoneally injected cannabinoids for 7 days,the intervention group(?)to the continuous intraperitoneal injection of cannabinoid receptor agonist GW405833 for 7 days,intervention group(?)to tramadol hydrochloride.Injections were intraperitoneally injected for 7 days.The pain test was performed on the intervention group(?),the intervention group(?),the intervention group(?)and the blank control group every 2 days.Four groups of rats were selected for prostate biopsy,pathological examination by HE staining,and the expression of CB2 receptor was detected from the protein and gene level by immunohistochemistry and Western blot,RT-PCR method.During the establishment of the EAP model,the pelvic pain test was performed in the control group and the EAP group on days 0,5,10,20,30,and 40 of Von-Frey pain measurement fibersResults:1.HE staining showed that the rats in the model group(EAP group)showed irregular changes in the prostate gland,infiltrating lymphocytes and neutrophils in the glandular interstitium,and congestion around the glands;there was no obvious inflammatory cells in the prostate of the blank control rats.Infiltration and congestion.2.Immunohistochemistry results:The expression of CB2 was detected in all groups of rat prostate cells,and the brown substance in rat prostate cells was CB2 receptor.CB2 was detected in successfully established rats.There are more cells than the blank control group.3.Western blot results:The expression of CB2 was detected in each intervention group and blank group,and there was a statistically significant difference in the expression of CB2 between the intervention group and the blank control group(P<0.01).CB2 was between the three intervention groups.The expression relationship was as follows:intervention group(?)>intervention group(?)>intervention group(?)>blank control group.4.RT-PCR results:Genes that express CB2 were detected in each intervention group and blank group.The expression of CB2 gene in the intervention group was significantly higher than that in the control group.The difference was statistically significant(P<0.01).Intervention group(?)>intervention group(?)>intervention group(?)>blank control group;Von-Frey pain test results showed that the response rate of pain in the intervention group rats was significantly higher than that in the control group(P<0.01);The results of Von-Frey pain test showed that the rats in the three intervention groups all reduced pain stimuli.The pelvic pain response in the intervention group(?,?)was significantly lower than that in the intervention group ?,and the difference was statistically significant(P<0.05).Conclusion(s):1.The rat model of experimental type ? prostatitis can be successfully constructed by multiple subcutaneous injections of Freund's adjuvant and prostate protein purification solution,supplemented by a double-immune adjuvant DPT vaccine.2.Von-Frey pain measurement fiber can be used to perform pelvic pain test in control group,EAP group and intervention group(?,?,?)rats;the test results are consistent with the pathological changes of prostate in each group.3.There are CB2 receptors in prostate tissue of normal rat and prostate tissue of EAP rats,and the number of CB2 receptors in EAP model group is significantly increased(P<0.05),indicating that CB2 receptor may be involved in The occurrence of chronic pelvic pain syndrome.4.Cannabinoid and cannabinoid receptor 2 agonists reduce pain in rats with chronic type ? prostatitis,compared to the traditional analgesic tramadol,its analgesic mechanism may be associated with cannabinoid receptors 2 is related,and its pathological changes are consistent with the Von-Frey fiber pain measurement results.
