Objective: To investigate the effect and significance of cox-2 inhibitors in the expression of ileum Acsl express of non-alcoholic fatty.Methods: 45 SD rats were randomly divided into normal control group and nimesulide group.All SD rats were fed in groups after one week of adaptive feeding and were fed with normal food to control group.At the end of the 11 th week,two rats were randomly selected from normal control group and NAFLD model group.Liver tissue HE staining and oil red O staining were used to determine whether the NAFLD model was successful.At the end of the 12 th week,total cholesterol of inferior vena cava was measured in each group.Total cholesterol(TC),triglyceride triglycer(Triglycer)were measured.The liver tissue of each group was stained by HE staining and oil red O staining to identify the degree of lipids degeneration and liver damage in each group.Real-time Quantitative PCR q PCR was used to detect the expression of COX-2and Acsl gene in ileum.Results: 1.NAFLD model group rats compared with normal control rats,increased level of serum TC,TG,(P < 0.05);The nimesulide group compared with NAFLD model group,the level of serum TC,TG all declined,(P < 0.05).2.oil red staining:the NAFLD model group have significantly steatotic(P<0.05),while in the nimesulide treatment group steatotic area is decreased significantly compared with NAFLD model group(P<0.05).3.HE staining: Compared with normal controls,NAFLD model group and nimesulide group all have liver fatty degeneration and liver structural damage,(P < 0.05).and compared with NAFLD model,nimesulide group liver steatosis reduced,no hepatic lobule formation.4.PCR detection results: NAFLD model group compared with normal control group,the expression of cox-2,Acsl3 and Acsl5 were increased(P < 0.05),the nimesulide group compared with NAFLD model group,cox-2,Acsl3,Acsl5 expression were significantly decreased(P < 0.05).Conclusion:cox-2 inhibitors can inhibit the progression of non-alcoholic fatty liver by down-regulating the expression of Acsl3 and Acsl5 in the ileum.
