Expression Of SIRT3 And FOXO3A In Osteoarthritic Cartilage And Its Biological Effects

Objective:To evaluate the correlation of SIRT3 and FOXO3 A in occurrence and development of OA and provide experimental evidence for studying the mechanism of OA.Method: The cartilage samples were obtained from 20 patients normal cartilage and 20 patients with osteoarthritis.Having HE staining and red O staining,according to the modified Mankin score standard,the osteoarthritis cartilage samples were classified into four groups.Immunohistochemistry method was used to explore expression of SIRT3 and FOXO3 A in cartilage,and to analyze its clinical significant.Results:1.the modified Mankin score standard :the modified Mankin score increased positively with degenerative grade.normal group is 0.65�0.48,mild degeneration group is 4.25�0.95,moderate degeneration group is 7.85�1.21 and severe degeneration group is 12.33�1.32.All groups are compared with each other that have statistically significant difference(P<0.05).2.The positive expression rates of SIRT3: SIRT3 was located in the cytoplasm of chondrocytes of normal cartilage and degenerative cartilage.The expression of SIRT3 decreased negatively with degenerative grade.normal group is(45.44�5.26)%,mild degeneration group is(38.38�5.04)%,moderate degeneration group is(30.72�6.33)% and severe degeneration group is(14.76�6.71)%.All groups are compared with each other that have statistically significant difference(P<0.05).3.The positive expression rates of FOXO3A: FOXO3 A was located in the chondrocytes of normal cartilage and degenerative cartilage.The expression of FOXO3 A decreased negatively with degenerative grade.normal group is(54.64�6.58)%,mild degeneration group is(44.51�6.07)%,moderate degeneration group is(27.29�5.43)% and severe degeneration group is(14.77�4.45)%.All groups are compared with each other that have statistically significant difference(P<0.05).4.the correlation of SIRT3 positive expression rates and the modified Mankin score standard: In degenerative cartilage,the expression of SIRT3 and the modified Mankin score standard was negative correlation and statistical significant(r=-0.914,P<0.05).5.the correlation of FOXO3 A positive expression rates and the modified Mankin score standard :In degenerative cartilage,the expression of FOXO3 A and the modified Mankin score standard was negative correlation and statistical significant(r=-0.943,P<0.05).Conclusion: Activity and expression of SIRT3 and FOXO3 A inosteoarthritic cartilage was significantly lower than normal cartilage,and negatively correlated with degenerative severity of cartilage.The expressive of SIRT3 and FOXO3 A were possibly related with the onset of osteoarthritic and its progress.
