Objective:To determine the expression of NEAT1,miR-101 and VEGF-C in bladder cancer tissues and adjacent tissues,to analyze the correlation between gene expression and age,gender,tumor grade,stage and metastasis and explore the potential mechanisms of invasion and metastasis of bladder cancer.Methods: First,we collected both cancer tissues and matched paracarcinoma tissues about 2cm away from the cancer from 30 bladder cancer patients in the First Affiliated hospital of South China University from January 2017 to December 2017.All the specimens were confirmed to be bladder urothelial carcinoma by pathological diagnosis after surgery,and the relevant pathological data were collected.Second,the data of age,gender,tumor stage,classification,and whether there was metastasis were statistically analyzed.Last,the expression and correlation analysis of bladder cancer tissues of 30 cases and adjacent tissues were analyzed by real-time quantitative PCRResults:1.Among 30 cases,23 cases were males and 7 cases were females.The age of male patients was 39 to 81 years,with an average age of 64.4 years.The age of female patients was 59-78 years,with an average age of 67.4 years.2.As to the expression of NEAT1 in bladder cancer tissues,there was significant(P < 0.05)difference between male and female,but there was no obvious correlation between expression level and age,tumor size,tumor number,clinical stage,differentiation degree and the presence of metastasis.As to the expression of miR-101 and VEGF-C in cancer tissues,there was no obvious correlation between the expression level and all of clinicopathological parameters mentioned above.3.The expression level of miR-101,NEAT1 and VEGF-C in the cancer tissue group was about 30%,4.15 times and 3.76 times of adjacent tissue,respectively.There was significant difference(P < 0.05)of the expression of these three genes between cancerous tissues and adjacent tissues.4.The correlation analysis shows that there was a correlation between the expression of NEAT1,miR-101,and VEGF-C.The expression of NEAT1 and miR-101,miR-101 and VEGF-C were both negatively correlated,while the expression of NEAT1 was positively correlated with VEGF-C(P<0.05).Conclusions:1.At the level of mRNA,compared to the expression in normal tissues,the expression of miR-101 in bladder cancer tissues was lower,while the expression of the NEAT1 and VEGF-C were higher.2.The expression of NEAT1 in bladder cancer tissue was correlated with gender,while miR-101 and VEGF-C had no significant correlation with above-mentioned clinicopathological parameters.3.NEAT1 probably promotes the expression of VEGF-C by inhibiting miR-101 expression,affecting the invasion and metastasis of bladder cancer.
