Effect Of Pokemon Silencing On Expression Of RFC4 In Breast Cancer Cell Lines And Its Correlation Analysis

Purpose To investigate the influence that the knockdown of pokemon produce on the expression of RFC4 in breast cancer cells,and to analyze the correlationship between Pokemon and RFC4.Methods Human breast cancer MDA-MB-231,HS578 T,MCF-7 and T-47 D celle lines were adopted as the experimental model,the sequence-specific pokemon si RNA was transfected into breast cancer cells,the influence of pokemon si RNA on the impact on the transcriptional level was detected by q RT-PCR,and the protein expression level of RFC4 was tested by the Western Blot.The expression rate of pokemon and RFC4 in 100 cases of non-specific invasive ductal carcinoma of the breast was detected by tissue microarray and immuohistochemistry,and the correlation between them was analyzed.Results Effect of Pokemon silencing on RFC4 in breast cancer cells: Pokemon Expression in four cell lines MDA-MB-231,HS578 T,MCF-7,and T-47 D Transiently Transfected with si RNA,m RNA Level of RFC4 Down-regulation indicates that silencing Pokemon from m RNA transcription level can inhibit the expression of RFC4.Pokemon was silenced in the four cell lines of MDA-MB-231,HS578 T,MCF-7,and T-47 D transiently by si RNA,and the protein level of RFC4 was down-regulated,indicating that silencing Pokemon from the protein level can inhibit RFC4 expression.To reveal the correlation between Pokemon and RFC4 expression in breast cancer tissues,the immunohistochemical analysis was made in the 100 cases of human breast cancer tissues(non-specific invasive ductal carcinoma).The expression of Pokemon and RFC4 was remarkably positively correlated.With the rise of Pokemon expression,the ratio of RFC4 expression level was elevated,the discrepancy was significant statistically.Conclusion 1)Silencing pokemon gene expression may restrain the levels of RFC4 in Breast cancer cell lines.2)The expression of Pokemon is positively correlated with RFC4 in breast cancer.
