Expression And Significance Of LncRNA ZEB1-AS1 In Ovarian Carcinoma

Objective: To study the expression and significance of lncRNA ZEB1-AS1 in ovarian carcinoma and its correlation with clinicopathologic features in ovarian carcinoma.Methods: The expression of ZEB1-AS1 were determined by the real time PCR.Student t test was used to exam expression of ZEB1-AS1 in ovarian carcinoma and noncancerous ovary tissue.The cutoff value of ZEB1-AS1 relative expression was determined by receiver operating characteristic curve(ROC).The chi-square test was used to exam the relationship between ZEB1-AS1 expression and clinicopathologic characteristics.The survival curves were estimated using the Kaplan-Meier method.The log-rank test was used to estimate the significance of the differences between the survival curves.A Cox proportional hazards analysis was performed to calculate the hazard ratio(HR)and the 95% confidence interval(CI)to evaluate the association between clinicopathologic characteristics and progression free survival(PFS)or overall survival time(OS).Results:(1)Real time PCR shown increased ZEB1-AS1 expression in ovarian carcinoma(52.99±43.90 vs 18.17±14.74,P<0.05).(2)ROC analysis shown cutoff was 21.68,with 91 cases high expression cases and 44 low expression cases.While the sensitivity is 67.41% and specificity is 66.67%.The high expression of ZEB1-AS1 correlated with high distant metastasis,more advanced FIGO stage and higher serum CA-125(p<0.05).(3)The ZEB1-AS1 high expression cohort of ovarian carcinoma favored recurrence than the low expression cohort(5-year recurrence rate: 91.21%(83/91)vs 47.73%(21/44),P<0.05).Cox regression analysis shown high ZEB1-AS1 expression was an independent factor for poor PFS.(4)The ZEB1-AS1 high expression cohort of ovarian carcinoma favored poor survival than the low expression cohort(5-year survival rate: 27.47%(25/91)vs 61.36%(27/44),P<0.05).Cox regression analysis shown high ZEB1-AS1 expression was an independent factor for poor OS.Conclusion:(1)ZEB1-AS1 testing predict the invasive behavior and higher clinical FIGO stage of ovarian carcinoma;(2)ZEB1-AS1 correlate with unfavorable prognosis.
