GelMA/MSCNs/BMP-2 Artificial Hydrogel Periosteum For Bone Regeneration

Objective: Prepare artificial periosteum based on Gelatin-MMA(Gel MA)membrane mixed with Mesoporous Calcium-Silica Nanoparticles(MCSNs)which loaded with BMP-2.The BMP-2 was released in sustained manner,and observed its bone formation ability.Methods: MCSNs were prepared with an improved templating agent method,and BMP-2 and absorbed into the nanoparticles in a freeze-dried way.Then the MCSNs-BMP-2 were incorporated into the Gel MA membrane to provide sustained release.The morphology and constitute of the MCSNs and the membrane were observed by SEM and FTIR.The mechanical character of the membrane were observed by a mechanical tester.The release manner of BMP-2 were determined by ELISA kits.The influence of cell adhesion and proliferation were observed by SEM and CCK-8.The osteoinductive capacity of composite scaffold was observed by alizarin red staining and alkaline phosphates(ALP).Composite scaffolds were implanted into the rat calvarial defects to test bone formation ability by histological and CImmunohistochemistry analysis.Results: At 1,10 and 28 days,21.2%±1.4%,58.5%±5.6% and 68.5%±6.7% of total BMP-2 were released from the scaffold.The SEM and CCK-8 proved that Gel MA membrane had good biocompatibility.The calcium content and ALP activity were improved by the hydrogel periosteum.In vivo study,scaffold with BMP-2 showed great bone value(BV/TV),and the newly formed bone could bridge the injury site.Conclusion: BMP-2 in hydrogel periosteum were released to mimic the process of bone healing.Hydrogel periosteum with BMP-2 had good biocompatibility and could promote bone formation.
