Objective: Through investigating the correlation between lacosamide and the expression of CRMP-2 and its phosphorylated protein in hippocampus of temporal lobe epilepsy rats,to clarify whether lacosamide can play an anti-epileptic effect by regulating their expression.Methods:The rats were randomly divided into three groups:(1)sham operation control group(control group),n=8;(2)epilepsy group(SE group),n=20;(3)epilepsy+lacosamide therapy group(SE+LCM group),n=20.SE group and SE+LCM group according to the time point divided into 3 d(n=4),7 d(n=8),14 d(n =4),28 d(n=4).The hippocampus of the rat was removed from the rat's head at 3 d,7 d,14 d and 28 d respectively.Then use western blotting technique monitor the expression of CRMP-2 and its phosphorylated protein in rats of each group was.In addition,the pathological damage of hippocampus was observed before and after of drug intervention of 7 d rats by making paraffin sections and HE stain.Results:(1)The number of chronic spontaneous seizures rats in SE+LCM group was less than that in SE group.(2)HE staining in optical microscope showedthat SE group and SE+LCM group rats CA1 regionpyramidal cells and DG regiongranular cells have a normal morphology,well-arranged and intact,and there was no obvious damage compared with Control group.However,the pyramidal cells in the CA3 region were obviously damaged.In SE group rats,the CA3 regionneurons from external ventral directionturned to ventromedial and continued to insert into dentate gyrusfell off and disappeared seriously,leaving only fragments and scattered normal cells.The neurons in the ca3 region of the rats in the left fragments and scattered normal cells.In SE+LCM group rats,the CA3 regionneurons only from external ventral directionturned to ventromedial parts fell off and disappeared seriously,while the rest of the neurons were relatively intact.(3)CRMP-2protein in hippocampus of SE group rats increased at first and then decreased at 3d,7d,14 d and 28 d,while the phosphorylation p Thr514 protein decreased first and then increased.(4)The expression of CRMP-2 protein in hippocampus of rats with temporal lobe epilepsy after lacosamide intervention was significantly lower than that of non-drug intervention group,and the expression of CRMP-2 phosphorylated p Thr514 protein was significantly higher than that of non-drug intervention group(P< 0.05).Conclusions:(1)Lacosamide has an anti-epileptic effect,and neuronal protectioneffect.(2)CRMP-2protein in hippocampus of SE group rats increased at first and then decreased at 3d,7d,14 d and 28 d,while the phosphorylation p Thr514 protein decreased first and then increased.(3)Lacosamide can inhibit the growth of neurite by inhibiting the expression of CRMP-2 and promoting its phosphorylated,thus play an anti-epileptic role.
