In Sleep and awakening are two different functional states of the human body.People can only perform various physical and mental activities in the state of awakening,and sleep can restore energy and physical strength.Therefore,adequate sleep is essential for promoting physical and mental health and ensuring normal physiological activities of the body.When sleep is deprived(or lost),there will be a strong rebound in sleep,indicating that sleep is regulated by homeostasis.Therefore,in sleep research,the sleep deprivation model is usually used to study the homeostasis mechanism of sleep.However,what parts of the brain are involved in homeostasis of sleep are not very clear.In this study,we used Wistar rats for 6 hours of sleep deprivation and 2 hours of sleep recovery after sleep deprivation.The changes in the expression of cytochrome oxidase(COX)in the related brain regions under the action of different sleep tension were observed by using EEG recording combined with histochemical staining and q-PCR detection.Objective to investigate the central location of endogenous regulation during sleep deprivation.The results showed that,compared with the normal group,the number of interconversion between the awakening and slow wave sleep was significantly reduced and the amount of sleep was increased in the electroencephalogram(EEG)record of the recovery group after sleep deprivation.The histochemical staining results showed that the sleep deprivation group and the sleep deprivation recovery group promoted the cells in the sleep system compared with the normal group.The activity of pigment oxidase(COX)increased and the activity of cytochrome oxidase(COX)in the awakening system decreased.The expression of melatonin type I receptor(MT1)in the habenular nucleus and pineal fruit showed that the MT1 gene expression in the sleep deprivation group and the recovery group after sleep deprivation in the pineal body was higher than that in the normal group;the rein nucleus slept in the habenular nucleus.The expression of MT1 gene in sleep deprivation group was higher than that in normal group,while MT1 gene expression in recovery group was lower than that in normal group after sleep deprivation.In this paper,the activity of sleep and awakening related brain regions and the regulation of MT1 in the habenular nucleus may be involved in sleep homeostasis in the habenular nucleus.The mechanism needs to be proved by further experiments.
