Laryngeal cancer is one of the most invasive head and neck malignant tumors.Its molecular mechanism is still unclear.Long non-coding RNA(lncRNA)can regulate the expression of tumor cell genes through a variety of mechanisms,including epigenetic mechanisms,and participate in the occurrence and development of various tumors.Insulin-like growth factor 1 receptor(IGF1R)regulates cell proliferation,differentiation and apoptosis through activation of multiple intracellular signaling pathways.IRAIN(IGFI receptor antisense imprinted noncoding RNA)is a lncRNA located in the IGF1 R gene region.It was first discovered in 2011 by Stanford University's tumor epigenetics laboratory.Objective:The purpose of this experiment is to explore whether IGF1 R antisense long-chain non-coding RNA-IRAIN affects the occurrence and development of laryngeal cancer,and provides a new molecular mechanism for the diagnosis,targeted therapy,gene therapy and prognosis of laryngeal cancer.Methods:6 samples of normal pharyngeal tissue and carcinoma tissues and para-carcinoma tissues from 37 patients with surgical treatment of laryngeal cancer were collected.ALL tissues were stored at-80 degress freezer.(1)In order to investigate the imprinting status of IRAIN in carcinoma tissues,para-carcinoma tissues and normal pharyngeal tissues,we extracted DNA and RNA from tissues and confirmed the status of imprinting of IRAIN allelic gene by PCR and DNA sequencing.(2)Quantitative Real-time PCR(qRT-PCR)was used to study the expression of IRAIN and IGF1 R in laryngeal carcinoma and paracancerous tissues.(3)Then the expression level of IRAIN and IGF1 R in laryngeal cancer and para-carcinoma tissues was analyzed,as well as the correlation between IRAIN and the clinical and pathological features.Results:IRAIN was diallelic exppression in normal pharyngeal tissues and adjacent laryngeal specimens,as well as in laryngeal cancer tissues.The expression level of IGF1 R is highly expressed in laryngeal cancer tissues compared with paracancerous tissues(P<0.05).The expression of IRAIN in laryngeal cancer tissues is lower than that in paracancerous tissues(P<0.05).The age,clinical classification,tumor differentiation,TNM stage and lymph node metastasis of patients with laryngeal cancer were not closely related to the expression of IRAIN(P>0.05).Conclusions:1.IRAIN was diallelic exppression in normal pharyngeal tissues and adjacent laryngeal specimens,so was in laryngeal cancer tissues.It suggested that the IRAIN allele was non-imprinted in normal pharyngeal tissues,laryngeal cancer tissues and paracancerous tissues.2.The expression level of IGF1 R is generally highly expressed in laryngeal cancer tissues compared with paracancerous tissues.It is suggested that IGF1 R may be related to the development of laryngeal cancer.3.The expression of IRAIN in laryngeal cancer tissues is lower than that in paracancerous tissues.This suggests that IRAIN may be involved in the occurrence of laryngeal cancer as a tumor suppressor.The age,clinical classification,tumor differentiation,TNM stage and lymph node metastasis of patients with laryngeal cancer were not closely related to the expression of IRAIN.4.The IGF1 R antisense long-chain non-coding RNA-IRAIN genomic imprinting may have tissue-specific expression patterns.
