Objective:Diabetic retinopathy(DR),is one of chief causes of irreversible blindness in working age population around the world.Laser photocoagulation?intravitreal injection of the antibodies against vascular endothelial growth factor(VEGF)?pars plana vitrectomy and so on are current popular therapeutic approaches for DR.They are invasive operations,which have some defects.So controlling the molecular switch in signaling pathways in gene levels provides us a new idea for the therapy of DR.The reverse transcription real-time fluorescence quantification technology analyses the expression of the serum mi RNA-19a?mi RNA-29 c and the ELISA meseasures serum VEGF content from patients of healthy subjects?T2DM patients and DR patients to search blood biomarkers for diagnosis of DR,and to provide theoretical basis for the further study of the role of mi RNA via cytokines such as VEGF in the pathological process of DR.Methods:A total of 30 patients with T2 DM were selected according to the standard of diagnosis of T2 DM and divided into two experimental groups based on their ophthalmoscope after mydriasis and fundus fiuorescein angiography results:T2DM group,n=15,8 males,7 females;DR group,n=15,7 males,8 females,and Control group-healhy subjects,n=15,6 males,9 females.Making use of Real-time PCR measures the serum expression levels of the mi RNA-19a?mi RNA-29 c and Using the ELISA meseasures VEGF level in groups,The correlation between the serum expression levels of mi RNA-19a?mi RNA-29 c and blood glucose and VEGF content was analysed with Pearson correlation analysis.Results:1.There is no significant difference in bloood pressure?BMI and LDLC in thress groups(p>0.05).Compared with Control group,Age?TG?TC and Hb A1 c were higher in the DR group(p<0.05),The Hb A1 c level in the DR group had greatly enhance contrasted with the T2 DM group(p<0.05),the differences were statistically significance.However,the rest indexs have no significant difference between the DR group and T2 DM group(p>0.05).In the DR group,HDL-C was inferior to Control group(p<0.05),which in the DR group has no significant difference with the T2 DM group.2.The serum VEGF expression in the Control,the T2 DM and the DR group gradually increased,serum VEGF increased 1.67 fold and 2.27 fold in the T2 DM and DR group than in the Control group(p<0.05),the differences were statistically significance.Serum VEGF increased 1.36 fold in the DR group than in the T2 DM group(p<0.05).Compared to healthy people,the expressions of mi RNA-19a?mi RNA-29 c were higher in the T2 DM group and the DR group(p<0.05),which in the DR group were superior to the T2 DM group(p<0.05),the differences were statistically significance.3.Pearson correlation analysis showed:Positive correlation between the expression level of mi R-29 c and Hb A1 c level was observed(p<0.05).With the increase of expression level of mi RNA-29 c,the Hb Alc level was raised.The expression levels of mi RNA-19 a and mi RNA-29 c were positively correlated with the serum VEGF content(p<0.05),which increased with the lift of the expression levels of mi RNA-19 a and mi RNA-29 c.Conclusion: 1.The expression of mi RNA-29 c positively correlates with Hb A1 c level,the expressions of mi RNA-19 a and mi RNA-29 c are positively associated with the serum VEGF content.2.Mi RNA-19a?mi RNA-29 c from DR patients may adjust glucose pathway?VEGF to participate in the pathogenesis of DR.3.Mi RNA-19 a and mi RNA-29 c may have potential to serve as blood index for diagnosis of DR.
