The Effect Of Endopensal Wolbachia On The Mitochondrial DNA Diversity Of Aedes Albopictus | | Posted on:2019-08-28 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q Q Liu | Full Text:PDF | | GTID:2394330545991431 | Subject:Pathogen Biology | | Abstract/Summary: | PDF Full Text Request | | ObjectiveThe regularity of Wolbachia infection in Aedes albopictus in 6 areas in Yangzhou was investigated.The genetic polymorphism of the cytochrome C oxidase subunit I gene of Aedes albopictus in 6 regions of Yangzhou was analyzed.The effects of endopensal Wolbachia on the mitochondrial DNA COI gene diversity of Aedes albopictus were analyzed.It lays the foundation for the control of mosquito vector and mosquito borne diseases based on Wolbachia characteristics in the future.Methods1.From July to October 2017,Aedes albopictus were collected from 6 regions of Yangzhou and the DNA of a single mosquito was extracted.The Wolbachia wsp gene of Aedes aegypti was detected by PCR and the infection rate was calculated.2.Using mitochondrial cytochrome C oxidase subunit I gene specific primers PCR amplification.The PCR products were sequenced by sequencers and the genetic diversity of the COI gene was analyzed by DnaSP5.1 software.3.The mitochondrial DNA diversity of Aedes albopictus populations infected with Wolbachia and without Wolbachia was analyzed using bioinformatics analysis methods.Results1.Detection results of Aedes albopictus infected with Wolbachia.The detection of Wolbachia infection in Aedes albopictus populations in 6 different areas of Yangzhou City had different degrees of infection and the infection rate was 27.5%-67.5%.The sequencing results of Wolbachia A large group and B large group showed that except for Aedes albopictus in Guangling area,there were Wolbachia superinfection in 5 other populations.The infection rate of only A group was 0~22.5%,the infection rate of only B group was 5%~17.5%,and the infection rate of infection A and B was 0~35%.2.Analysis of genetic polymorphisms of the cytochrome C oxidase subunit I gene of Aedes albopictusThe A+T content of Aedes albopictus COI gene(67.93%)was significantly higher than that of C+G(32.07%),nucleotide diversity(Pi)was 0.28190,haplotype polymorphism(Hd)was 0.933.3.Evolutionary tree based on COI gene sequence.The evolutionary tree based on NJ method showed that Aedes albopictus collected from Hanjiang and Jiangdu sampling areas were clustered into a small branch,and then became a group with Guangling district and Yizheng district.The Aedes albopictus collected from Gaoyou and Baoying counties were individually gathered into one branch.There were differences in the mitochondrial COI gene of Aedes albopictus at different sampling points in Yangzhou.4.The effect of Wolbachia on the mitochondrial DNA COI gene diversity of Aedes albopictus.Except for Guangling population,haplotype diversity(Hd),nucleotide diversity number(Pi)and nucleotide mean number(K)of 5 other populations without Wolbachia infection were all higher than those of Wolbachia infection.Conclusions1.The infection of Wolbachia in the population of Aedes albopictus in Yangzhou showed diversity and superinfection.2.The COI gene of Aedes albopictus in 6 different geographical populations in Yangzhou has a certain degree of gene exchange.3.The nucleotide diversity and haplotype diversity of the mitochondrial DNA COI gene of Aedes albopictus population infected with Wolbachia were lower than that of the Aedes albopictus population without Wolbachia.Wolbachia has a certain influence on the diversity of mitochondrial DNA in the host. | | Keywords/Search Tags: | Wolbachia, Aedes albopictus, Mitochondrial DNA, Cytochrome C oxidase subunit I gene, Nucleotide diversity, Haplotype diversity | PDF Full Text Request | Related items |
