BackgroundCurrently,the treatment of Parkinson's disease mostly depends on drugs,which is not ideal.The effects of deep brain stimulation(DBS)techniques,or it's on neural circuits,remain to be studied.Optogenetics can be used to treat Parkinson's disease by using the specific expression of the photoreceptor gene,controlling specific neurons or neural pathways as needed,and human intervention of cells.On the basis of existing theory,we design experiments independently,collect and analyze the EEG signals before and after the treatment,and then evaluate the effect of the treatment to provide a certain technical support for the use of optogenetics technology in Parkinson's disease.Objective1.Preparating of Parkinson's disease rat model by stereotaxic injection,and selecting a variety of model evaluation method:behavior(apomorphine induced,treadmill test),EEG signal,morphological analysis.2.Building a therapeutic scheme based on light stimulation.3.Study the EEG signal before and after treatment,obtain the rule of characteristic value and evaluate the therapeutic effect.Methods1.Using stereotaxic instrument to find the disease-causing regions,and injecting rotenone to prepare of Parkinson's disease rat model.After a week,inducting and carrying on treadmill training,then collecting EEG signal and completing HE dyeing for the success rate of model preparation.2.Selecting successful model for injecting of the virus,and suturing after surgery,after virus expression selected parts of the rat randomly for frozen section to see the virus expression under fluorescence microscope.3.Implanting optical electrode:4×4 optical electrode was prepared to implant corresponding brain region.4.The EEG signal of Parkinson's rats was recorded before the treatment,and the sampling frequency was 1000 Hz,twice daily and 10 days in a row.5.Light stimulation treatment:treatment frequency is 25 Hz,treatment intensity is 10 mw/mm~2,and recording signal changes after treatment,the sampling frequency is 1000 Hz,twice daily,10 consecutive days.6.The analysis of EEG signals before and after treatment,after pre-processing,the local field potential changes and LZ complexity were calculated.Results1.Model behavior results:rats appeared the contralateral rotation,the bow back,the stiff tail,tremor.The experiment of touch number on running machine after modeling is obvious much less,and the startup time of fore limb is longer.HE dyeing showed that neuron necrosis and the number decrease.2.After the injection of virus,red neurons were observed under the microscope to prove the expression.3.After the treatment of light stimulation,the energy value and LZ complexity of the collected local field potentials of model tended to the blank rats,which met the expected therapeutic results.Conclusion1.Using stereotaxic injection of rotenone to preparate of Parkinson's disease rat model,rats appeared to the lateral movement disorders,morphological experiments demonstrate that the behavior result is because of neuronal necrosis caused by the damaged side.2.Treatment with a certain wavelength,frequency and intensity light can improve the disorder of Parkinson's EEG.From the microscopic perspective,it is because the light stimulation changes the firing patterns of neurons.
