Objective 1.To analyze the change of insulin sensitivity after spinal cord injury(SCI)by observing the change of the blood glucose,the glucose tolerance,the phosphorylated S6 protein(pS6)and the c-Fos expression in hypothalamic.2.To preliminary understand the function of PI3K pathway in insulin resistance or diabetes mellitus type 2 following SCI.Methods A total of 120 rats were randomly divided into four groups:the sham group with common feedstuff,the sham group with "3H"-hyperpressure hyperlipemia and hyperglycemia feedstuff,the SCI group with common feedstuff,the SCI group with hyper:pressure hyperlipemia and hyperglycemia feedstuff,each group had 30 rats."3H" feedstuff was used to induce insulin resistance and T2DM.All of them drank water freely daily.Animal model with incomplete SCI was made by modified Allen's method.The sham group performed the same operation without injury of spinal cord.After 8 weeks of feeding,the rats in each group were tested for weight,blood glucose and glucose tolerance,and the insulin resistance of rats in different groups was observed.Then the rats were executed and the spinal and hypothalamus were separated,then the immunofluorescence detection of pS6 and c-Fos positive neurons in hypothalamus were observed.In order to understanding preliminarily the effects of the PI3K signaling pathway on hypothalamus in SCI.Results The weight increased less in the SCI group rats than the sham group,but the blood glucose increased more.The glucose tolerance was lower than the sham group.And the difference was statistically significant.The three observation indexes had more obvious differences between in the "3H" groups.On the other hand,the greater expression of c-Fos in the hypothalamus was found in SCI group with common feedstuff than sham group,and it was even greater in SCI group with "3H" feedstuff.And the lower expression of pS6 in the hypothalamus was found in SCI group with common feedstuff than sham group,and it was even lower in SCI group with "3H" feedstuff.Conclusion The blood glucose rose and the glucose tolerance reduced of the SCI rats in the study.And it proved that SCI is an excess risk of T2DM.The injury in spinal cord had a vital effect on the expression of pS6 and c-Fos in hypothalamus under the condition of feeding with "3H" feedstuff.SCI effects on neuronal responses in the hypothalamus which cause the lower expression of pS6 but the greater of c-Fos,may resulting in inhibiting PI3K signaling,decreasing the inflammatory reflex and increasing the releasing of many inflammatory mediators,ultimately leading to insulin resistance and T2DM.
