ObjectiveThe Wistar rat model was infected by Respiratory Syncytial Virus(RSV)to observe the appearance,mental state,activity,lung histomorphology and pathological changes,lung index,Interferon-?(IFN-?),Interleukin-10(IL-10),hormone metabolism and their correlations were investigated in order to investigate the changes of appearance and the reproduction of virus in RSV-infected rats,Lung tissue injury,pathological changes,inflammatory mediators and hormone levels in order to explore the pathogenesis and mechanism of disease development after RSV infection and provide a theoretical basis for early clinical diagnosis and treatment.MethodsWistar rats were divided into RSV group and control group by randomized grouping method.RSV group was inoculated intranasally with RSV Long strain virus solution.By observing the RSV infection at different time points rat appearance,mental status,activity,lung morphology and pathological changes,lung index,viral load and other indicators analysis and evaluation.The levels of IFN-? and IL-10 were detected by Elisa method and the related hormones were detected by liquid chromatography-mass spectrometry(LC-MS).The correlation between cytokines and hormone metabolism at different time points after RSV infection was analyzed.Result:1.On the 3rd and 7th day after RSV infection,the appearance,mental status,activity and growth status of rats in RSV group were significantly higher than those in the control group,and were progressive increase(P <0.05).The viral load of lung tissue of rats in RSV group increased significantly on the third day(2.97 ± 0.07)and decreased on the seventh day(1.26 ± 0.04).From the appearance of lung tissue,The appearance color gradually changed from light red to dark red.The smooth and inflammatory cells infiltration.moist lungs gradually became uneven and the elasticity gradually changed from good to bad.On the lung pathology,RSV rats lung tissue alveolar normal tissue disappeared,alveolar wall telangiectasia congestion,consolidation of alveolar wall thickened,edema,peripheral blood vessels infiltration by a large number of2.On the 3rd and 7th day after RSV infection,compared with the 0th day of infection,the expression of IFN-?protein in rat serum first increased and then decreased significantly(P <0.05).IL-10 cytokines were significantly increased(P <0.05),and significantly decreased on the 7th day compared with the 3rd day after infection(P <0.05).3.On the 3rd and 7th day after RSV infection,compared with the 0th day of infection,the metabolites 11-deoxycortisol,21-deoxycortisol and tetrahydrocortisone Tetrahydrocorticosterone)were significantly increased first,and then decreased significantly with statistical significance(P <0.05).4.The expression of Cortexolone,21-Deoxycortisol,Tetrahydrocorticosterone and IFN-?,IL-10 cytokines in rat serum after RSV infection.The levels were positively correlated with statistical significance(P <0.05).Conclusions1.The general status of RSV rats changed after infection,lung index,viral load measurement,lung appearance,lung histopathology under the light microscope to provide an objective basis for the identification of RSV infection model.RSV infecting Rat model is successful.2.After infection with RSV,the related cytokines changed,Th1-type cytokines decreased and Th2-type cytokines increased.3.RSV infection in rat serum hormone metabolites change significantly,which is involved in the RSV virus infection.4.After infection with RSV,the related cytokines in the body are correlated with the changes of hormone metabolism,which play a role in the occurrence and development of the disease.
