ObjectivesTo study the therapeutic effect of Baitouwong jiajianfang enema on ulcerative colitis in rats and explore its possible mechanism.MethodsThe 60 male SD rats were randomly divided into 6 groups,with 10 in each group.The groups were setted: control group,model group,Baitouwong jiajianfang low concentration group,middle concentration group and high concentration group,and salazosulfapyridine solution group.Except for the control group,the ulcerative colitis model of SD rats was prepared by the combination of trinitrobenzene sulfonic acid/ethanol.After the success of the model,the drug enema was given to the corresponding drug enema,and the concentration of the drug in the low,middle and high concentration group was 0.125g/ml,0.25g/ml and 0.5g/ml.In the group of salazosulfapyridine solution,the solution was used to fill the enema with the mixture of sulfazosulfopyridine enteric solution(concentration of 50 mg/ml).The control group and the UC model group were treated with distilled water enema,each group was 2 ml each time,one time a day for consecutive fifteen days.After fifteen days,we observed the general conditions of the rats,the disease activity index,the colonic mucosa damage index,the histopathological score,and the apoptosis index,serum IL-6 and IL-10 in rats were detected by Enzyme-linked immunosorbent assay.The expression of MUC2 in rat colon was detected by Immunohistochemistry.Results1.Compared with the control group,model group rats disease activity index,the colonic mucosa damage index,histopathological score,apoptosis index,serum IL-6expression increased(P < 0.05),serum IL-10,MUC2 expression were lower(P < 0.05).2.Compared with model group,Baitouwong jiajianfang the low,middle and high concentration group and sulfazosulfonate solution disease activity index,the colonic mucosa damage index,histopathological score,apoptosis index,the level of serum IL-6 significantly decreased(P < 0.05),the level of serum IL-10,MUC2 expression were significantly increased(P < 0.05).3.Compared with Baitouwong jiajianfang middle concentration group,Baitouwong jiajianfang low concentration the contents of serum IL-10,reduce MUC2 expression,disease activity index,the colonic mucosa damage index,histopathological score,apoptosis index,serum IL-6 levels increase,the difference was statistically significant(P < 0.05);Baitouwong jiajianfang high concentration group had no significant difference.Conclusion1.Baitouwong jiajianfang can reduce the degree of tissue inflammation,and it has a good anti-inflammatory effect on ulcerative colitis,and can regulate the secretion of cytokines IL-6 and IL-10.2.The expression of MUC2 in the colon mucosal tissue can be increased by the Baitouwong jiajianfang,which can protect and repair the colonic mucosal barrier.3.Baitouwong jiajianfang is possible to improve the expression of MUC2 and IL-10 and reduce the expression of IL-6 and play a therapeutic role in ulcerative colitis by raising the expression of MUC2 and IL-10.4.The change of IL-6,IL-10 level in serum and MUC2 expression reminder that the concentration and therapeutic effect of the addition and subtraction of the Baitouwong jiajianfang are related to the therapeutic effect.
