Abjective:By analyzed the impact of age and sex of the healthy donors on the content of CD34~+cells in the bone marrow,it provides an important reference for the bone marrow collection of donors in clinical.Method:A total of 96 healthy donors who donated bone marrow stem cell(BMSC)in the First Affiliated Hospital of Guangxi Medical University from December 2013 to January 2018 were analyzed.According to the age and the distribution characteristics of the data,they were divided into 5 groups:the age?28d(newborn),the 28d<age?3yr(infant),the 3yr<age?10yr,the 10yr<age<18yr and the?18yr groups,respectively.The leukocyte count was used to calculate the nucleated cell count(NCC).NCC was measured by blood cell analyzer,and the percentage of CD34~+cells was measured by flow cytometer,and the CD34~+cell count was calculated.The relationship between age,sex and NCC,the percentage of CD34~+cells and CD34~+cell count were analyzed.Results:1.Among the 96 healthy donors,the median percentage of CD34~+cells in bone marrow collections was 1.43(0.55,3.15).Among them,the percentage of CD34~+cells in this 5 groups were 2.40(1.53,4.00),3.30(2.33,3.65),1.10(0.68,1.44),0.50(0.28,0.70)and 0.28(0.24,0.50),respectively.The difference between the newborn group and the infant group was not statistically significant(P=1.000),and the last 3 groups were all significantly lower than the infant group(P=0.005,P<0.001,P<0.001,respectively).2.NCC(×10~9/L)was 21.13(16.27,23.64)in the newborn group and 22.74(14.06,28.00)in the infant group,there was no significant difference between this two groups(P=0.383);There were 54.40(38.76,71.17),52.97(40.08,76.00)and 50.77(43.19291.97)respectively in the latter 3 groups,and there was no significant difference between the groups(P=0.922).3.The CD34~+cell count was 4.67(2.61,6.89)in the newborn group and6.37(3.92,9.39)in the infant group,there was no significant difference between this two groups(P=1.000).And the infant group was still higher than that of2.38(1.10,4.18)in the 10yr<age<18yr group and 1.83(1.26,3.91)in the?18yr group(P=0.034,P=0.016,respectively).4.The percentage of CD34~+cells and the CD34~+cell count both have negative correlations with age(r=0.638,P<0.001 and r=0.223,P=0.029,respectively).When male and female donors were evaluated separately,the percentage and count of CD34~+cells were still negatively correlated with age in this two groups,(r=-0.667,P<0.001 and r=-0.245,P=0.089 in male group;r=-0.578,P<0.001 and r=-0.170,P=0.255 in female group,respectively).5.The percentage of CD34~+cells,NCC(×10~9/L),and CD34~+cell count(×10~8/L)were 1.80(0.55,3.20),25.68(19.60,50.71),4.15(2.38,6.96)for males and 1.10(0.53,3.10),34(22.73,54.95),4.93(1.99,8.75)for females,respectively,and these values were not statistically significant(P=0.150,P=0.312,respectively).Conclusions1.The percentage of CD34~+cells and CD34~+cell count of bone marrow were negatively correlated with age,and had no correlation with sex;2.The percentage of CD34~+cells of bone marrow in newborns and infants are significantly higher than that of adults;3.With the same number of CD34~+cells,the younger donors can reduce bone marrow collection,of which newborns and infants can be reduced by 2.57and 3.51 times compared with adults.
