Hepatocellular carcinoma(hepatocellular,carcinoma,HCC),is one of the most common malignant tumor.The existence of Strong concealment,Progress is rapid,easy occurrence of intra hepatic metastasis,distant metastasis,radical resection rate is low and the recurrence rate of higher treatment difficulty.A lot of Epidemiological datas show that Diabetes with the incidence of HCC development has a very close relationship.A recent study found that,RAGE play a role in HCC carcinogenesis factors,its downstream cytokines MKK4,MKK7 and JNK1 played in human hepatocellular carcinoma role is not yet clear.Up to now,has yet to see RAGE,MKK4,MKK7,JNK1 expression level in the relevant reports of Diabetic patients with Hepatic Carcinoma(referred to as DMHC),therefore the issue to be discussed.Objective:In order to further explain the expression of RAGE,MKK4,MKK7 and JNK1 protein in DMHC,and its expression differences with the above factors in HCC,and the clinicopathological features were analyzed.Methods:Western blot was applied to detect the expression of RAGE?MKK7?MKK7 and JNK1 in cancer tissues of 18 cases of DMHC and 30 cases of HCC and the corresponding adjacent tissues.Results:? The level of RAGE?MKK7?p-MKK7 and JNK1 proteins was higher in cancer tissues of both HCC and DMHC group than their corresponding adjacent tissues,while MKK4 and p-MKK4 expression was low(all P<0.05).Compared with the HCC group,MKK7,p-MKK7 and JNK1 expression in cancer tissues of the DMHC group was enhanced(all P<0.05),while the expression of RAGE,MKK4 and p-MKK4 proteins wasn't significantly changed(all P>0.05).? In the HCC groups,the expression of RAGE was increased in poorly differentiated cancer tissues as compared to highly differentiated cancer tissues(P<0.05).In the HCC and DMHC groups,the expression of MKK7,p-MKK7 and JNK1 was increased in poorly differentiated cancer tissues as compared to highly differentiated cancer tissues(all P<0.01).? In the DMHC group,the expression MKK4 in cancer tissues of patients with cancer matastasis was significantly decreased(P<0.05).? The level of RAGE,MKK4,MKK7 and JNK1 in these two groups wasn't significantly correlated with HbsAg and the size of HCC(all P>0.05).The level of RAGE,MKK7 and JNK1 in these two groups wasn't significantly correlated with matastasis of HCC(all P>0.05).? The level of RAGE was both positively correlated with the expression of MKK7 and JNK1 in cancer tissues of the HCC and DMHC groups(all P<0.05).Conclusions:? The increase of RAGE expression may be involved in the occurrence and development of HCC in T2DM.? The expression of MKK7 and JNK1 in DMHC group was significantly up-regulated,and the expression of MKK7 and JNK1 was more significant than that of HCC group,and the expression of MKK7 and JNKlmight promote the progress of HCC.? The reduction of MKK4 expression may have the effect of promoting tumor metastasis in DMHC group.? The expression of MKK7 and JNK1 may be related to the differentiation of liver cancer cells in DMHC group.
