Objective:To establish a rat model of chronic inflammatory induced by lipopolysaccharide?LPS?.To explore the regulating effect of Laminarin L01 on inflammation related factors induced by LPS and its related mechanisms,from that to open up new ideas for the prevention and treatment of cardiovascular diseases.Methods:?1?Establishment of chronic inflammatory rat model:A small dose of LPS(0.4 mg·kg-1)was injected via tail vein once a week for 4 weeks.The inflammatory reaction was evaluated by leukocyte count and interleukin-6in serum.?2?Experimental animal grouping and administration:SD rats were randomly divided into 6 groups.In addition to the blank control group,only sterile saline was injected.The other 5 groups were respectively given intraperitoneal injection of dexamethasone(10 mg·kg-1)and L01 high,medium and low dose(50,30,10 mg·kg-1)the day after the first injection of LPS.LPS group was injected with the same volume of aseptic saline once a day for 4weeks.?3?Index detection:Anus temperature of rats was measured;After24hour of the last administration,the number of whole WBC was counted;IL-6in serum was measured by ELISA,the expression of HSP70 and TNF-?mRNA were detected by RT-PCR and the related products were examined by density and half-quantitative analysis after electrophoresis.Detection of protein expression of HSP70,TNF-?and VEGF by immunohistochemistry.Results:?1?The number of WBC and the serum level of IL-6 increased significantly in the 1thh week of LPS injection?vs.blank control group,P<0.05?,and the injection of LPS in the 2th,3thh and 4thh week maintained that at a high level.There was no statistically significant difference among the 1th,2thh 3thh and4thh week groups.?2?LPS increased WBC counts and serum IL-6 levels in rats?vs.blank control group,P<0.01?.The changes can be antagonized by the therapeutic administration of different concentrations of L01 for four weeks.?3?L01 decreased HSP70 and TNF-?mRNA expression.?4?L01 decreased aorta potein expression of HSP70,TNF-?and VEGF.Conclusion:Laminarin L01 could decrease the expression of inflammatory factors and leukocytes count in whole blood in chronic inflammatory rats,decrease serum content of IL-6,HSP70 and TNF-?,down-regulate the expression of HSP70 and TNF-?mRNA,as well as,protein expression of HSP70,TNF-and VEGF.The results suggest that the potential anti-inflammatory effect of Laminarin L01.
