The Influence On Metastasis Of Bladder Cancer T24 Cells By Interfered With RhoGDI2 Gene

Objective: To study the effect of interference with RhoGDI2 on the metastasis of bladder cancer in vitro and in vivo and that provide useful information for the study of the treatment in bladder cancer metastasis.Methods: 1.Design and synthesize lentiviral vectors which contain interfered with RhoGDI2 gene,and use western blotting technique to test RhoGDI2 gene in the differences among T24-RhoGDI2-RNAi,T24-RFP and T24 RhoGDI2 gene.2.T24-RhoGDI2-RNAi and its control cell strains were cultivated in animals as well.3.T24-RhoGDI2-RNAi and its changes in invasion and metaptosis of control cell strains in vitro and in vivo.4.To set up a pulmonary metastasis model by intravenous injection of T24-RhoGDI2-RNAi and its control cell strains at caudal of the nude mice.5.The comparisons of growing time of the tumor,the metastasis rate and the survival of the model.Results: 1.Have set up lentivirus vector that is interfered RhoGDI2 gene.Western blotting technique have tested in RhoGDI2 protein content of T24-RhoGDI2-RNAi is significantly lower than that of T24-RFP and T24 group(P < 0.05).2.In the experiment of invasion and metaptosis,the cell number of invasion and metastasis of Group T24-RhoGDI2-RNAi is much more than that of Group T24-RFP and Group T24(P < 0.05).Another comparison of control group,it is no statistical that the Group T24-RFP and Group T24 about the number of cells for invasion and metastasis have any differences.3.Group T24-RhoGDI2-RNAi has the lowest survival in nude mice pulmonary metastasis model in vitro.4.The gender of the nude mice survival analysis showed no statistical significance.(P > 0.05)Conclusion: Interference of RhoGDI2 gene helps to promote the invasion and metastasis of T24 bladder cancer,and reduce the survival time of the animals.It is showed that interference of RhoGDI2 gene help to promote metastasis of bladder cancer,to affect the survival prognosis of the nude mice.RhoGDI2 gene could be a prohibitive metastasis factor at the early stage of bladder cancer.RhoGDI2 gene is closely related to tumor size,differentiation,tumor stage,metastasis and vascular invasion.
