Objective:Through the observation of the expression levels of CCR7 protein and the Th1/Th2 balance in asthma model rats exposed to cigarette smoke,which is under intervention of Bailing Capsule,the research aimed to explore an immunological mechanism for cigarette smoke aggravate asthma and the Bailing Capsule treatment airways inflammation,put forward experimental basis for the treatment of asthma.Methods:Forty healthy male SD rats were randomly divided into blank control group(A),asthma group(B),smoking asthma group(C),Bailing Capsule asthma group(D),Bailing Capsule smoking asthma group(E),each group of eight.The rats of latter four groups were sensitized with the ovalbumin 1ml injected intraperitoneally on its first day and eighth day and challenged by inhaling 2% ovalbumin 60 ml placed in transparent airtight container to establish the asthma model,one time a day,every time 30 minutes,six times a week,eight weeks a total.Form the challenged,the smoking asthma group and Bailing Capsule smoking asthma group rats were put in the self-made fumigating box(the size is 60cm*60cm*50cm)to smoke passively,ten cigarettes one day,60 minutes one time,eight weeks in all.Form the challenged,Bailing Capsule asthma group and Bailing Capsule smoking asthma group were fed Bailing Capsule 30 min before each challenging,the dose of 0.945g/kg.The blank control group give saline 2 ml instead of ovalbumin intraperitoneal injection,in the 15 th day 60 ml saline instead of 2% ovalbumin aerosol inhalation,the other remaining conditions is kept the same.In 24 hours after the last inhalation,intraperitoneal injection of chloral hydrate anesthesia,the rats were killed,take the lung tissue complete,the lung tissue were embedded in paraffin and sliced.The expression of CCR7 protein in airway were detected using immunohistochemistry,IFN-?and IL-4 were both quantitatively analyzed in serum of rats by Enzyme-Linked Immuno Sorbent Assay(ELISA).Results:(1)The expression of airway CCR7 protein of group B(0.357±0.046)and group C(0.433±0.053)were significantly higher than group A(0.123±0.027),the difference was statistically significant(all P<0.05);Group C was higher than group B,the difference statistically significant(P<0.05);Group D(0.238±0.054)lower than group B,statistically significant difference(P<0.05);Group E(0.300±0.046)lower than group C,statistically significant difference(P < 0.05).(2)The level of serum IFN-? of group B(52.83±13.28)and group C(31.29±7.76)were significantly lower than group A(134.27±17.67),the difference was statistically significant(all P<0.05);Group C was lower than group B,the difference statistically significant(P<0.05);Group D(96.45±12.25)higher than group B,statistically significant difference(P<0.05);Group E(67.12±9.10)higher than group C,statistically significant difference(P < 0.05).(3)The level of serum IL-4 of group B(62.77±9.28)and group C(83.26±10.21)were significantly higher than group A(32.14±8.95),the difference was statistically significant(all P<0.05);Group C was higher than group B,the difference statistically significant(P<0.05);Group D(42.25±8.75)lower than group B,statistically significant difference(P<0.05);Group E(52.65±10.90)lower than group C,statistically significant difference(P < 0.05).Conclusion:(1)CCR7 may be related to airway inflammation of asthma rats.(2)Cigarette smoke can increase the expression level of airway CCR7 protein,affect the Th1/Th2 balance,aggravate the airway inflammation.(3)The Bailing Capsule can reduce the expression level of airway CCR7 protein,controlling the Th1/Th2 imbalance,reduce the airway inflammation.(4)Based on conventional treatment,combined use of immunomodulatory drugs,may increase the efficacy of asthma in clinical.
